| Objective:The study aims to explore the mechanism between the transformation of AMCC and asthma through taking unilateral adrenalectomy as a deficiency of kidney-yang modle and the interference of kidney-tonifying recipe on asthmatic rats.Methods:Fifty six rats were divided into seven groups in random (n=8):asthmatic rats (Asthma), unilateral adrenalectomy asthmatic rats (Surgery-asthma), kidney-tonifying asthmatic rats (KTA), unilateral adrenalectomy kidney-tonifying asthmatic rats (Surgery-KTA), dexamethasone asthmatic rats (DA), unilateral adrenalectomy dexamethasone asthmatic rats (Surgery-DA) and control rats (Control). Asthma, Surgery-asthma, KTA, Surgery-KTA, DA and Surgery-DA rats were sensitized and challenged with ovalbumin (OVA) rather than Control. The pathological changes of adrenal medulla were observed by microscopy. EPI and its rate-limiting enzyme, Phenylethanolamine N-methyl transferase (PNMT), were measured with immunohistochemisty. Rho GDP dissociation inhibitorsα(Rho GDâ… Î±) and Peripherin, relating to reconfiguration of cytoskeleton in AMCC, were also detected in each group.Results:1. Compared with asthma group, asthmatic rats showed a significantly thicker airway wall, infiltration of eosinophil and lymphocyte surrounding airway, vacuolar degeneration and increased lipid in AMCC, while the degree of damage in surgery asthmatic rats was more serious. Compared with asthmatic rats, the degree of damage in KTA and DA rats showed been improved. Compared with surgery asthmatic rats, surgrey-KTA and surgery-DA rats appeared less lesions. Electron microscope showed a regular morphology of AMCC in control rats which contained a clear structure of mitochondria and abundant chromaffin granules. Some manifestations of lesions, swelled cytoplasm and mitochondria, decreased chromaffin granules, presented in asthmatic rats:. In addition to the performance above in asthmatic rats, contents of AMCC disintegrated, which exhibited apoptosis in surgery asthmatic rats. At the same time, axon appeared in AMCC of asthmatic rats, which was deemed as an interesting sign. Compared with asthmatic rats, the degree of damage in KTA and DA rats was meliorated. Compared with surgery asthmatic rats, surgrey-KTA and surgery-DA rats appeared less lesions.2. The expressions of EPI and PNMT in asthma rats were more significantly decreased than those in control rats (P<0.05). Compared with asthmatic rats, a further decline was viewed in surgery asthma rats (P<0.05), but there was an enhancement of those in KTA and DA rats (P<0.05). Compared with surgery asthmatic rats, surgery-KTA and surgery-DA rats showed an increasing expression of EPI and PNMT (P<0.05).3.The Rho GDIa expression in the asthmatic rats was less than control rats (P<0.05). Compared with asthmatic rats, surgery asthmatic rats had a further decrease (P<0.05), and there was an enhancement of Rho GDIa in KTA and DA rats (P<0.05). Compared with surgery asthmatic rats, surgery-KTA and surgery-DA rats showed an increasing expression (P<0.05), while the expression of Peripherin was contrary to Rho GDIa (P<0.05).Conclusion:1.Unilateral adrenalectomy could aggravate the alteration of the structure and function of AMCC and promote the transformation from AMCC to neuron in asthmatic rats.2. Kidney-tonifying recipe could inhibit the transformation from AMCC to neuron in asthmatic rats.3. Rho GDIa and Peripherin may play an important role in the transformation of AMCC. |
