Objective: To investigate the effect of recombinant human endostatin on hypertrophic scar(HS) of rabbit ear and the expression of vascular endothelial growth factor ( VEGF) and tissue inhibitor of metalloproteinase-1(TIMP-1) expression.Methods: 16 New Zealand rabbits were used to establish hypertrophic scar model.The rabbits were randomly divided into two groups, which were experimental group and control group ( 8 each group ), after hypertrophic scar model was established successfully. Rabbits in experiment group were injected with recombinant human endostatin, and others in control group were injected with physiological saline.The changes of HS include color, texture and thickness were observed. The HS harvested and detected by Hematoxylin and Eosin (HE) as a routine examination and by SP immunohistochemistry and RT-PCR to observe the expression of VEGF and TIMP-1.Results: The color of scar was weaker , texture was sofer and thickness was thiner in experimental group compared with those in control group . In HE dyeing, dermis was thiner , capillary was smaller in experimental group than those in control group. Hyperplasia index(HI) of scars in experimental group and control group were 1.37±0.21 and 2.65±0.21 respectively, and the statistical difference was significant(P<0.01). The mRNA and protein expression level of VEGF and TIMP-1 in experimental group were significantly lower than those in control group (P<0.05 ).Conclusion: Recombinant human endostatin can inhibit hyperplasia of hypertrophic scar of rabbit ear, which maybe through the way of suppressing the expression of VEGF and TIMP-1.
