Objective: To investigate global protein expression profiles in the rabbits retinas of normal and proliferative vitreoretinopathy (PVR) by proteomic analysis.Methods: PVR was induced by vitrectomy and creating retinal breaks in rabbits first,then all rabbits were raised for 8 weeks. Age-matched untreated rats served as controls. Retinal protein expression profiling was carried out by two-dimensional gel electrophoresis (2-DE). Proteins were identified by high-performance liquid chromatography and tandem mass spectrometry using a chip cube nano-flow system (HPLC-Chip–MS/MS system) (Agilent). Alpha-crystallin B chain - Oryctolagus cuniculu (CRYAB) was validated by western blot.Results: In experimental group, PVR–B and PVR-C was induced in 8 weeks after rabbits bad been performed vitrectomy and creating retinal breaks surgury. In PVR group, levels of two proteins were altered, as revealed by 2-DE and HPLC-CHIP-MS/MS: Alpha-crystallin B Chain (CRYAB) and Alpha-crystallin A Chain (CRYAA), and both upregulated more than 3-fold, P < 0.05. The expression level of CRYAB was increased significantly in PVR retinas compared with the normal retinas and was consistent with that in 2-DE data.Conclusion: CRYAB may play an important role in the pathogenesis of PVR. |
