Cerebral ischemia-reperfusion injury may affect the turnover ofdisease in patients, even threaten their lives, but its pathogenesis has notbeen very clear now, so the research of its mechanism and the curativedevelopment were paid much more attention. In the study, the model ofcerebral ischemia-reperfusion was set up in order to observe the effect ofKangNaoYe on the contents of SOD, MDA, NO, NOS and amino acids ofserum and brain tissue in rats during cerebral ischemia-reperfusion to studythe neuroprotection and its protection mechanism of Kangnaoye.284 rats were randomly divided into 6 groups: sham-operation group,ischemia-reperfusion group, KangNaoYe group with high dose(28.60g·kg-1·d-1), KangNaoYe group with medium dose(14.30 g·kg-1·d-1),KangNaoYe group with low dose(7.15 g·kg-1·d-1), Edaravone group(10.00mg·kg-1·d-1). There were 54 rats in each group. The models of cerebralischemia-reperfusion were established in rats by the methods of middlecerebral artery occlusion(MCAO). The rats were separated into groupssuch as 1h, 3h, 6h, 12h, 24h, 48h after 2 hours of operation. The animalswere killed to the deadline of the schedule, observed the pathologicalchange after HE Staining and measured the volume of infarction of thebrain by the method of TTC. The contents of SOD, MDA, NO, NOS weremeasured by spectrophptometer. The contents of Asp, Glu, Gly,γ-GABA inserum and brain tissue were detected by reversed-phase High PerformanceLiquid Chromatography with pre-column derivatization respectively.Compared with shamed group, the symptom of Neurological function,the volun of cerebral infarction and the distention of neuron were severity, the activity of SOD was reduced and the content of MDA, NO, NOS, Asp,Glu, Gly,γ-GABA and the ratio of Gly/γ-GABA was improved inischemia-reperfusion group. There were significant difference(P<0.05).KangNaoYe and Edaravone could relieve the symptom of Neurologicalfunction, reduce the volum of cerebral infarction, improve the swelling anddistension of neuron and interstitial edema to different degrees, elevate thecontent of SOD, Gly andγ-GABA, decrease the content of MDA, NO,NOS, Asp and Glu. Compared with ischemia-reperfusion group, there weresignificant difference(P<0.05). Compared with Edaravone group, thesymptom of Neurological function, the volume of infarction of the brainand the pathological change were relieved in KangNaoYe group withmedium and high dose. The content of SOD, Gly andγ-GABA wasimproved and the content of MDA, NO, NOS, Asp ,Glu and the ratio ofGly/γ-GABA was cut down. There were significant difference(P<0.05).There were not significant difference between KangNaoYe group withmedium and high dose (P<0.05).The injury of ischemia-reperfusion can increase the release of freeradical and excitatory amino acids in the serum and brain tissue, so thecontent of inhibitory amino acids is not enough compared with EAA.KangNaoYe can relieve the ischemia-reperfusion injury of brain andaugment the protection of nerve tissue, inhibit the injuries of free radicaland accelerate the banlance of the amino acid metabolism in blood andbrain tissue.
