Objective:To study the protective effect and mechanism of Jinqiaomai(JQM) on lung and heart injury induced by Klebsiella pneumonia in rats .Methods:Establish Klebsiella pneumonia model by instillation of bacteria drops through trachea; The male SD rats were randomly divided into control group, model group, JQM (6, 3, 1.5g/kg) three groups and levofloxacin (25mg/kg) group; Observe the general state of health of the rats; Extract the heart,serum and lung tissue 6 days later; WBC and PMN in peripheral blood were counted ; The content of IL-1β, IL-6, IL-8, TNF-α, ICAM-1, INF-γin serum and lung tissue homogenate and TNF-α, ICAM-1 in heart tissue homogenate were measured by radioimmunoassay and Elisa; TNF-α, ICAM-1, NF-κB p65 protein expressions in lung and TNF-α, ICAM-1 protein expressions in heart were measured by immunohistochemistry; MIP-2 mRNA expressions in lung was detected by in situ hybridization; TLR2/4 mRNA and MyD88 mRNA expression in lung were detected by RT-PCR; IκB-αexpression lung was detected by Western Blot; The organizational structure and ultrastructural organizational change of lung and heart were observed .Results:The various doses of JQM can significantly improve the general state of health of the rats; Compared with the control group, the counts of WBC and PMN in peripheral blood of the model group were markedly increase(P<0.01), the contents of IL-1β, IL-6, IL-8, TNF-α, ICAM-1 and INF-γin serum were significantly increase (P<0.05 or P<0.01). Compared with the model group, the counts of WBC and PMN in peripheral blood of JQM (6, 3, 1.5g/kg) three groups were markedly decrease (P<0.05 or P<0.01), the contents of IL-6, IL-8, ICAM-1 and INF-γin serum of JQM (6, 3, 1.5g/kg) three groups were significantly reduced (P<0.05 or P<0.01) ;Compared with the control group, the rats of model group had obvious lung injury, the contents of IL-1β,IL-8,ICAM-1 and INF-γin lung tissue homogenate of model group were significantly increase (P<0.05 or P<0.01), the contents of TNF-α,ICAM-1,NF-κB p65 and MIP-2mRNA in lung of model group significantly increase (P<0.05 or P<0.01), the contents of TLR2/4 mRNA,MyD88 mRNA and IκB-αin lung of model group significantly increase (P<0.05 or P<0.01). Compared with the model group, Jinqiaomai (6, 3, 1.5g/kg) three groups had less injury, the expressions of IL-1βand ICAM-1 in lung tissue homogenate of Jinqiaomai (6, 3, 1.5g/kg) three groups were significantly lower than those in the control group (P<0.05 or P<0.01), the expressions of INF-γin lung tissue homogenate of Jinqiaomai (6g/kg) group were significantly decrease (P<0.05), the expressions of TNF-α,ICAM-1,NF-κB p65 and MIP-2 mRNA in lung of JQM (6, 3, 1.5g/kg) three groups were significantly decrease, the contents of TLR2/4 mRNA,MyD88 mRNA and IκB-αin lung of JQM (6, 3) groups were significantly reduced (P<0.05 or P<0.01);Compared with the control group, the rats of model group had obvious heart injury, the contents of TNF-αand ICAM-1 in heart of model group were significantly increase (P<0.05 or P<0.01). Compared with the model group, the contents of TNF-αand ICAM-1 in heart of Jinqiaomai(6, 3, 1.5g/kg) three groups were significantly lower than those of model group(P<0.05 or P<0.01). Conclusion:JQM has significant protection on Klebsiella pneumonia in rats, which may be related to the inhibition of Toll-like receptor MyD88 dependent on channel in lung and the content of IL-1β,TNF-α, IL-8,ICAM-1and INF-γin serum and lung.JQM has significant protection on heart injury induced by Klebsiella pneumonia in rats, which may be related to the inhibition of TNF-αand ICAM-1 in heart .
