Objective: Currently the malignant tumor scares the mankind's health and life, and also it trends that more and more young people got it, which makes people feel so terrified to hear that. What is worse, the number of patients increases gradually. And colorectal cancer is the most common among malignant tumor of digestive tract. Studies have shown that colorectal cancer was the results of colon epithelial cells affected by both genetic and environmental factors. So the only way to cure cancer thoroughly is through the research of gene. Malignant tumor is a difficult problem worldwide and there is no good way to eradicate diverts and relapses completely. Now we can only deal with it by operation, chemotherapy, radiation treatment and some other ways. However, the ways mentioned above can only kill the remained cancer cells and make relapse also metastasis deferred, ending with the results that people give up treatment because of the serious side-effect. Until now, the pathogenesis of colorectal cancer is still unclear. With the extensive development of molecular biology and gene academic, people get to know more about the pathogenesis of tumor and genetic. Nowadays it is generally agreed that colorectal cancer is a matter of multi-gene, multi-step complex process involving multiple oncogene activation and tumor suppressor gene inactivation. The research discovers that the tie rectum cancer is the result that is acted by syntheses such as heredity and environment etc. to cause the gene mutate on skin cell of bowel, so cans by the root cure of cancer only from the gene quest.The tumor is a hardly world proplem,CDX2 is new-found cadual type homebox gene, and its main function is to regulate usual development and differentiation of epithelium on normal bowel. If the expression of CDX2 reduces or loses, a series of gene regulation will turn abnormal, and at last ending with tumor. The gene KISS-1was separated from tumor cells of melanoma , by Lee and named as gene KISS-1. The combination of albumin coded by gene and acceptor GPR54 can repress the invading and transferring of tumor cells.The study is to investigate the relationship between the expression of tumor-associated gene CDX2,KISS-1 in Colorectal carcinoma expression,and discovered the processing of the cancer,s developing , transferring and carcinoma ventriculi. it Provided the evidence of diagnoising, curing and prognosticing aspectsMethods: The expression of CDX2 and KISS-1 were detected using immunohistochemistry SP method, and the expression was concerning to 45 cases of colorectal cancer,the corresponding proximal adjacent tissues(5cm from the cancer tissue) and 20 cases of normal colorectal tissues and they were compressive analyzed acrroding to the patient's age, sex, location, infiltration depth,differentiation degree,lymph node metastasis and Dukes stage, using SPSS13.0 statistical software for statistical data processing, x2 test count data(P<0.05 significant difference) which analysed the relationship between the expression of both CDX2 and KISS-1 in colorectal cancer by using Spearman rank correlation.Results:(1)The positive expression of CDX2 was 66.7% in the colorectal cancer tissues, it was 88.9% in proximal cancer tissues and 95% in normal colorectal tissues. The expression of CDX2 in cancer tissues was quitely lower than the proximal adjacent tissues and normal colorectal tissues (P<0.05, significant difference), and the expression of CDX2 in the proximal adjacent tissue was quitely lower than normal colorectal tissues (P<0.05,significant difference).(2) The positive expression of KISS-1 was 35.6% in the colorectal cancer tissues, 44.4% in proximal cancer tissues and 80% in the normal colorectal tissues. The expression of KISS-1 in cancer tissues was quitely lower than the proximal adjacent tissues and normal colorectal tissues (P<0.05, significant difference), and the expression in the proximal adjacent tissue was quitely lower than normal colorectal tissues (P<0.05,significant difference). (3) The expression of CDX2 and KISS-1 in colorectal cancer had close relationship with tumor invasion depth , Duke's stage ,lymph node metastasis(P<0.05). CDX2 had close relationship with the degree of differentiation of cancer tissue (P<0.05), while KISS-1 had no relationship with the degree of differentiation of cancer tissue (P>0.05). its expression had close relationship with the degree of differentiation cancer tissue, invasion depth, lymph node metastasis (P<0.05). but had no obvious relationship with the patient's age , the position of the cancer and gender.Conclusions:(1) the expression of CDX2 in colorectal cancer had closely relationship with tumor invasion depth, Dukes stage, The expression of CDX2 in infiltrating muscularis of the colorectal cancer was lower than in Baptist serosa of the colorectal cancer.The expression of CDX2 in poorly and no differentiated colorectal cancer was lower than in differentiated and well-differentiated colorectal cancer. The expression of CDX2 in Dukes staging of C+D stage colorectal cancer was lower than in Dukes staging of A+B stage colorectal cancer, the degree of differentiation of cancer tissues and lymph nod metastasis. The expression of CDX2 in colorectal cancer with lymph node metastasis was lower than in colorectal cancer without lymph node metastasis. (2) the expression of KISS-1 in colorectal cancer had closely relationship with tumor invasion depth,Dukes stage and lymph nod metastasis. The expression of KISS-1 had no obvious relationship with tumor differentiation The expression of KISS-1 in infiltrating muscularis of the colorectal cancer was lower than in Baptist serosa of the colorectal cancer. The expression of KISS-1in Dukes staging of C+D stage colorectal cancer was lower than in Dukes staging of A+B stage colorectal cancer, The expression of KISS-1 in colorectal cancer with lymph node metastasis was lower than in colorectal cancer without lymph node metastasis. (3)The expression of CDX2 and KISS-1 in colorectal tissues positively correlated(r=0.384,P<0.05). So CDX2 and KISS-1 gene are all tumor suppressor gene,and the two have obvious synergistic action in the generating, developing and transferring process of carcinoma ventriculi. Therefore,the joint evaluation of the genes has more important significance in diagnoising, curing and prognosticing aspects.
