Objective:Injury of tendons has long been a common clinic problem in the department of Hand Surgery . Autograft tendon transplantation, transposition, extension and replacement with autograft fascia for tendon defects are commonly used, but the source of autograft tendon is limited. Multitude tendon defects in the clinical treatment is still an urgent problem to solve. As the study and preparation of allograft tendon blaze a new trail for tendon defects reconstruction with the following advantages: wide source, convenient drawing, shortened operation time, reduced new injury upon the patients and functional influences upon the donor site,and allogenic tendon transplantation has been the main method for tendon defects. Now there is more use of deep-frozen treatment on tendon allograft, however, although treated by the deep-frozen, 16 months after the operation of frozen allograft tendon transplantation, specific organization phase dissolve antibodies can be still checked out, and inflence tendon transplant effect.Both tendon graft or tendon repair, the adhesion of tendon is inevitable after the operation, which affects the limb function seriously. Tripterygium wilfordii hookf has the functions of anti-inflammation, anti-immune, antisterility, antitumor, and antimicrobial activity. Amniotic membranes can promote the growth of the load cell, so it usually act as the carrier of cell growth and proliferation. It also can prevent tendon adhesion. The amniotic membrane is a semi-permeable membrane whose unique structure can prevent certain substances from passing through. Amniotic membrane can package xenogeneic cells or tissues (such as allogeneic islets and other tissues and organs) in its sac to avoid immune rejection. This experiment research the anti-rejection effect of triptolide in tendon allograft transplantation and the anti-adhesion of amniotic membranes which does no increase the rejection in tendon allograft transplantation.Methods: Establish Leghorn chickens'third toes allograft tendon transplantation model. The experimental groups were randomly divided into experimental group I, experimental group II, experimental groupâ…¢, and a control group, 32 Leghorn chickens in each group. Do tendon allograft transplantation in the control group. After three weeks and six weeks, collect peripheral blood from eight Leghorn chickens,then do the streaming cytological examination cd4, cd8 and cd4 / cd8 and biomechanical test in Pmax and W0; observe it around electron microscope after two weeks and four weeks, and around light microscope after one week and three weeks. In Groupâ… and Groupâ…¢, add triptolide in the feed, while Groupâ…¡and Groupâ…¢pack amniotic membranes at the tendon cut ends, three and six weeks after the tendon transplantation, do biomechanical test and streaming cytological examination cd4+,cd8+T lymphocytes.Results: Analyzing the streaming cytological examination cd4, cd8 and cd4 / cd8 of three-week and six-week by statistical software, peripheral blood contents less cd4,cd8 in Groupâ… and Groupâ…¢than in Groupâ…¡and the control group, and the ratio of cd4/cd8 increased; there is no evident difference between Groupâ…¡and the control group. Biomechanical test shows that Groupâ…¡and Groupâ…¢is better than other two groups, and Groupâ… is better than the control group. Electron microscopy and light microscopy can show the tendon healing in Groupâ… and Groupâ…¢is better than the other two groups.Conclusion: Triptolide can prevent the rejection in tendon allograft transplantation, and reduce the content of cd4, cd8 effectively. Amniotic membranes have the function of anti adhesion and promote tendon healing in tendon allograft transplantation without new rejection.
