Objective: To investigate the expression of extracellular matrix metalloproteinase inducer (EMMPRIN/CD147) and matrix metalloproteinases-9 (MMP-9) in human laryngeal carcinoma, the relationship between the expression of CD147 and MMP-9 in tissues with the concentration of plasma CD147 and MMP-9 in the corresponding patients, as well as the relationship between the expression of CD147 and the concentration of plasma MMP-9 with the invasion and metastasis of laryngeal carcinoma.Methods: (1) The expression of CD147 and MMP-9 was analyzed semi-quantitatively by immunohistochemical staining in laryngeal carcinoma and control group samples. (2)The total CD147 and MMP-9 in plasma (free and complex form of CD147 and MMP-9) were measured by enzyme linked immunosorbent assay (ELISA).Results: (1) The positive rate of CD147 was 83.3% in laryngeal carcinoma, which was higher than in laryngeal polyp (33.3%) or in para-carcinoma tissue (16.7%).(2)The positive rate of MMP-9 was 72.2% in laryngeal carcinoma, which was higher than in laryngeal polyp (13.3%) or in para-carcinoma tissue (5.6%). (3)The expression rate of CD147 and MMP-9 in laryngeal carcinoma was both 72.2%,in which positive correlation was found (rs=0.721,P=0.000).(4)There was a positive correlation between the expression of CD147 in tissues and the concentration of plasma CD147 in the corresponding patients(rs=0.921,P=0.000).The more intensive immunohistochemical staining,the higher the average concentration of plasma CD147 was. (5)There was a positive correlation between the expression of MMP-9 in tissues and the concentration of plasma MMP-9 in the corresponding patients(rs=0.914,P=0.000).The more intensive immunohistochemical staining,the higher the average concentration of plasma MMP-9 was. (6) There was a positive correlation between the concentration of plasma CD147 and MMP-9 in laryngeal carcinoma patients(rs=0.974,P=0.000).The higher the average concentration of plasma CD147,the higher the average concentration of plasma MMP-9 was. (7)The concentration of plasma MMP-9 in Laryngeal carcinoma was higher than in laryngeal papilloma, laryngeal polyp, laryngitis, or normal people (P<0.01). (8) The expression of CD147 was related with histopathological grade, clinical stage and whether there was lymph node metastasis (P<0.05). (9)The concentration of plasma MMP-9 was related with age, clinical stage and whether there was lymph node metastasis (P<0.05). With the change of clinical stage(Ⅰ/Ⅱ→Ⅲ/Ⅳ) and lymph node metastasis (without metastasis→Transfer), the concentration of plasma MMP-9 increased, but decreased with increasing age. But there were no correlation between the concentration of plasma MMP-9 with sex, primary tumor site, histopathological grade, T stage respectively.Conclusion: CD147 and MMP-9 have a higher expression and intimate expressive relationship in laryngeal carcinoma. CD147 has relationship with the occurrence, development, invasion, and metastasis of laryngeal carcinoma. The concentration of plasma MMP-9 is higher in laryngeal carcinoma. The abundance of plasma MMP-9 might originate from the excessive expression of tissue MMP-9,which in a certain indicate whether there is lymph node metastasis.
