| ObjectiveTo investigate the expression and clinicopatholagical significance of farnesoid X receptor (FXR) and nuclear factor-KB(NF-tcB) protein in hepatocellular carcinoma (Hepatocellular carcinoma, HCC).MethodsHE staining,Collagen staining and Reticular fiber staining were used to determine the inflamrnatian grade and fibrosis stage of the 56 liver cancer patients para-cancerous liver tissues,far-cancer liver tissues.Furthermore,the protein expression of FXR and NF-tcB were detected by immunohistochemical staining method (two-step chemistry) and Western Blot method in HCC tissues,para-can cerous liver tissues, far-cancer liver tissues and 16 normal liver tissues, then the difference of them and the correlation with clinicopathological features werecompared and analysised.ResultsThe positive rate(expression) of FXR protein in HCC tissues, para-cancerous liver tissues,far-cancer liver tissues,and normal liver tissues was 8.9%(0.7110.39),4$.2%(0.8310.42),85.7%(1.2010.88),87.5%(1.0610.2),respectively.The expressi ons of FXR protein in HCC tissues was significantly lower than in para-cancerous liver tissues,the expressions of FXR protein in para-cancerous liver tissues was lower than in far-cancer liver tissues,and normal liver tissues (p<0.05).The positive rate(expression) of NF-xB protein was 82.1%(0.2110.11),60.7%(0.1510.07},28.6%(0.1110.04),12.5%(0.1210) in HCC tissues,para-cancerous liver tissues,far-cancer liver tissues and normal liver tissues,respectively.The expressions of NF-xB protein in HCC tissues was significantly higher than in para-cancerous liver tissues,the expressions of NF-xB protein in para-cancerous liver tissues was higherthan in far-cancer liver tissues,and normal liver tissues(P<0.05).The correlation coefficient of FXR and NF-KB protein in HCC tissues,para-cancerous liver tissues,far-cancer liver tissues,and nortxtal liver tissues was -0.967,-0.873,-0.858 and -0.842,P<0.O1,respectively.The correlation coefficient of FXR protein and the differentiation of HCC,the copies of HBV-DNA,the grades of liver inflammation and the stages of liver fibrosis was -0.766,-0.784,-0.926 and -0.798,P<0.05。Conclusion1. The expression of FXR protein was lower but the expression of NF-KB protein was higher, the expression of FXR protein and NF-r}B were negative correlation in all liver tissues.2. the expressions of FXR protein and the differentiation of HCC,the copies of HBV DNA,the grades of liver inflammation,and the stages of liver fibrosis were negative correlation.There was not statistical difference in other clinicopathological features. |
