Objective Observation of Total alkaloid of Peganum harmai on murine S180 sarcoma cells and H22 hepatoma cells'inhibition in vivo and in vitro. Observation of Total alkaloid of Peganum harmai on human hepatoma cells HepG2's proliferative inhibition in vitro.Methods 1. Total alkaloid of Peganum harmai on murine S180 sarcoma cells and H22 hepatoma cells'inhibition in vivo and in vitro:To establish transplanted tumor mouse model through inoculating tumor cells subcutaneously with Kunming mice, observed the effct on weight of tumor-bearing mice, spleen index and thymus index at a concentration of Total alkaloid of Peganum harmai at 60 mg/kg, 30 mg/kg, 15mg/kg by a continuous infusion for 10 days;2. Total alkaloid of Peganum harmai on human hepatoma cells HepG2's proliferative inhibition in vitro: MTT assay was used at different concentrations of Total alkaloid of Peganum harmai at different time points of the murine S180 sarcoma cells, H22 hepatoma cells and human hepatoma cells HepG2's proliferative inhibition, to calculate inhibition rate and half inhibitory concentration (IC50), the morphologic features of cells were observed under inverted microscope.Results 1. Total alkaloid of Peganum harmai on murine S180 sarcoma cells and H22 hepatoma cells'inhibition in vivo and in vitro: Total alkaloid of Peganum harmai can inhibit the growth of mouse S180 sarcoma cells,It has a relatively good inhibition when inhibition rate is between 9.30%~56.13%, total alkaloid of Peganum harmai's concentration is at 30 mg/kg;When inhibition rate is between 12.98%~36.59%, total alkaloid of Peganum harmai's concentration is at 60 mg/kg,It has a relatively good inhibition on liver cancer cells in mice H22 tumor, but has no significant impact on the spleen index and thymus index; 2. Total alkaloid of Peganum harmai on human hepatoma cells HepG2's proliferative inhibition in vitro: Total alkaloid of Peganum harmai on S180, H22 and the proliferation of HepG2 cells were significantly inhibited, the relative inhibitory effect was best at 2000μg/mL, 1000μg/mL, and the inhibition was enhanced with extension of time, it was relatively best up to 72h. IC50 of the S180, H22 and HepG2 hepatoma cells at Different time points is 2435.74μg/mL~299.99μg/mL, 1263.12μg/mL~306.34μg/mL, 549.08μg/mL~27.20μg/mL respectively;HepG2 cells in the negative control group was observed under inverted microscope ,which the adherent cells grow well,spindle-shaped, adherent HepG2 cells in the drug delivery group are less, most were round suspended in solution.Conclusions 1. The Total alkaloid of Peganum harmai has a good proliferation on murine S180 sarcoma cells and H22 hepatoma cells in vivo and no damage to the immune organs;2.The Total alkaloid of Peganum harmai has significant impact on proliferative proliferation of murine S180 sarcoma cells, H22 hepatoma cells and human hepatoma cells(HepG2) in vitro. It indicates that the Total alkaloid of Peganum harmai has better anti-tumor effect, it is worth studying further and provide experimental basis for developping into a new anti-cancer herbs. |
