| Mouth and throat disease is a common disease of human, which is mainly due to infections caused by bacteria, measured by research, the main infective bacteria is Streptococcus, Staphylococcus, and Streptococcus pneumonia and so on. Although the oral cavity and throat diseases will not immediately lead to human's death, because of its diversity of diseases, easy to relapse, difficult to cure, and many of the complications caused by them and bringing a lot of inconvenience and trouble to human's life. Therefore, the diseases of mouth and throat have been concerned all the time.Currently, the human anti-bacteria and viruses rely mainly on antibiotics, but because of antibiotic treatment will have many adverse consequences, including the side effects of the coordination mechanism, risk of drug resistance increased caused by bacteria, viruses and so on. In recent years, people began to study with immunological method, and found that using IgY technology can be produced antibodies for drug-resistant, which can treat infections caused by bacteria or viral disease. It is safety and no residue, no resistance, low cost and other advantages, and is a promising alternative to antibiotic.In this study, we prepared specific IgY against Streptococcus pneumonia and MG-hemolytic streptococcus, and detected its biological properties by SDS-PAGE electrophoresis, Coomassie brilliant blue staining method, bacteriostatic test, ELISA, cross-reaction, and studied heat stability, acid stability, pepsin stability, trypsin stability of IgY.The results showed that we can obtained high purity of IgY by the acetic acid-sodium acetate buffer dilution, polyethylene glycol precipitation and salt precipitation method; indirect enzyme-linked immunosorbent assay (ELISA) showed that yolk antibody is produced in hens 10 d after the first immunization, the titer gradually increased 20 d after the first immunization, and the higher titer produced in 40-80 d after the initial immunization, the highest titer of IgY against Streptococcus pneumonia can reach up 1:5120, the highest titer of IgY against MG-hemolytic streptococcus can reach 1:2560 maximum; antibacterial experiment showed that specific IgY against Streptococcus pneumonia, MG-hemolytic streptococcus can significantly inhibit the bacterial, the inhibition rates can reach to 90%; cross-reaction of the specific IgY can specifically bind to its bacteria antigen.In summary, in this study we successfully prepared with high content, high purity and specific IgY against Streptococcus pneumonia, MG-hemolytic streptococcus; antibacterial experimental shows that IgY can effectively combine with its bacteria antigens, and has good inhibition of bacteria, which laid the foundation for the prevention and treatment of pharyngitis. |
