The Study Of Effects Of Liu-wei Di-huang Pills And Four Original Constituents On The Proliferation And Differentation Of Rat Preadipocyte

Objective:Effects of Liu-wei Di-huang Pills containing the ratio of loganin,morroniside,peoniflorin,paeonol to blood on the proliferation and differentiation of rat preadipocyte.By Liu-wei Di-huang Pills with drug-containing serum and plasma on rat preadipocyte proliferation and differentiation of the various time points, compared with plasma and serum containing drug at all time points between efficacy of experimental results.Methods:Rat preadipocytes were cultured originally, then above-mentioned four original constituents according to 4 factors and 3 levels to make L9 (34) orthogonal test and set the various time points of Medicated serum and plasma to culture preadipocytes The difference of absorbancy were compared in the proliferation rates of rat Preadipocyte were determined by MTT method, and the differentiation was tested by Oil Red O staining.Results:1 The combination of the four original constituents of liu-wei Di-huang pills promote the proliferation and inhibit differentiation of rat preadipocytes. Affect factor of the order is D>C>A>B,and the best choice is AlB3C1D1.That is Loganin with high concentrations 8.5μg/ml; Morroniside with Low concentration 1.25μg/ml; Peoniflorin with high concentrations 8.0μg/ml; Paeonol with high concentrations 9.0μg/ml. 2 the Medicated serum and Medicated plasma of liu-wei Di-huang pills have a significant effect on proliferation of rat preadipocytes,and the most obvious time point group is 60th Minute. Statistical analys shows the the difference of two delivery methods of Medicated serum and Medicated plasma was not significant (P>0.05).The difference among control group and 5,10,15,30,45,60,120,180,240,360min time group was significant (P <0.05).The difference among 60 min group and control group, 5,10,15,30,45,120,180,240,360min time group was significant(P<0.01) liu-wei Di-huang pills can promote the proliferation of rat preadipocytes.It shows there was no difference between Medicated serum and Medicated plasma of liu-wei Di-huang pills The most Medicated serum and Medicated plasma significant effect proliferation of rat preadipocytes is after intragastric administration rat 60 minutes. Medicated serum of liu-wei Di-huang pills inhibit differentiation of rat preadipocytes, the most significant effect is rat Blood sampling time point at 60 minutes.Conclusion:1 The combination of the four original constituents of liu-wei Di-huang pills promote the proliferation and inhibit accumulation of Cytoplasmic fat differentiation process of rat preadipocytes.A significant effect on Loganin, morroniside,Paeonol with high dose,but the Peoniflorin with concentrations make no sence. The Orthogonal design combination of Loganin;Morroniside; Peoniflorin; Paeonol Reprensent the overall regulatory role of synergy and efficiency.2 The Medicated serum and Medicated plasma of liu-wei Di-huang pills at Different blood sampling time points is 60th Minute group.it shows a significant effect on proliferation or differentiation of rat preadipocytes This conclusion is most guidance value for clinical applications of liu-wei Di-huang pills.3 In the In vitro experiments study of preadipocytes of liu-wei Di-huang pills, Serum-Pharmacology and Plasma pharmacology result no difference.
