Aim:The objiectives of this study were to investigate the inhibitory effects of resveratrol on HCN4 Participation Coding Hyperpolarization Activated Cyclic Nucleotide Gated Cation Channel Current(If), and trying to identify its role site,explore its mechanism. Methods: HCN4 plasmid DNA extracted, And transcription into mRNA in vitro,then HCN4 expressed channels in oocytes of Xenopus laevis and studied by using standard two-microelectrode voltage-clamp technique.Results: HCN4 is inhibitoryed in a concentration- and voltage- dependent manner by resveratrol (half-maximal inhibition concentration[IC50]=83.75 umol/L), and the steady state activation and inactivation parameters are shifted to more positive values in the presence of 160umol/l resveratrol. State-dependent test revealed voltage-dependent If blockade occurred rapidly. In this experimental, we were introduced isoproterenol, We were found that Isoproterenol has positive function for HCN4 participation of the super polarization activation code of cationic channel current,it can make the current express increase by more than 50%,then Join 160umol/L resveratrol we found resveratrol also has strong inhibition to the enhance current.Conclusion: resveratrol has inhibitory effect to HCN4 Participation Coding Hyperpolarization Activated Cyclic Nucleotide Gated Cation Channel Current(If). And the effect is to effect on channels directly. Resveratrol on isoproterenol enhanced participation encoding HCN4 hyperpolarization activated cation channel currents with a significant inhibitory effect. It prompted that resveratrol may had some clinical value of the treatment of arrhythmias induced by isoproterenol.
