Objective To investigate the effect of diallyl disulfide (DADS) on cell proliferation of human leukaemia K562 cell, and the role of DADS on the expressions of Bcl-2,Bax and Survivin.Methods MTT was used to test the inhibitory effects of DADS on proliferation in K562 cells. Flow cytometry with FITC labeled annexin V staining was used to determine the percentage of apoptotic cells. Western Blotting was used to analyze the expression of Bcl-2/Bax and Survivin. And the mRNA expression of Bcl-2/Bax and Survivin were detected by RT-PCR.Results DADS significantly inhibited proliferation of K562 cells in a concentration dependent manner. After treatment with 20μM,40μM,80μM and 160μM DADS for 24h, K562 cells growth inhibit rates were 5.55%,14.81%,33.33%,48.15%. Data also demonstrated that DADS significantly induced increase of the percentage of apoptotic cells (P<0.05), in a time- and concentration-dependent manner. The results also displayed that DADS significantly inhibited the expression of Bcl-2, enhanced the expression of Bax, inhibited the protein expression of Survivin but not mRNA in the K562 cells.Conclusion DADS inhibits the proliferation of K562 cells and effectively induces apoptosis of K562 cells, which mechanism may be associated with downregulations of Bcl-2,Survivin and upregulation of Bax.
