Objective:1. Analyze the difference of caries sensitivity and dentin sensitivity of nasopharyngeal carcinoma patients before and after radiotherapy treatment.2. The purpose of the present study was to analysis the change of salivary protein total amount and several key proteins content and quantified to detect the content of lysozyme and lactoferrin for nasopharyngeal carcinoma patients before and after radiotherapy, explore its relationship with radiation caries.Method1.Resazurin disc method to detect caries activity.2. VAS method to measure dentin sensitivity.3.BCA method to determine total content of saliva protein, SDS-PAGE gel electrophoresis to separate saliva protein components, use Quantity One professional gel images analysis software to scan and analyze the image, measured the IOD value of the target band.4.Westerblotting to detect the content of lysozyme and lactoferrin in saliva protein, chemiluminescence(CIA) to obtain the X-ray film images of target band, Quantity One image analysis software to specialize scanning images, measure the IOD values of target band. Analysis the difference of the content of lactoferrin and lysozyme in saliva before and after radiotherapy, and discuss the relationship among lactoferrin, lysozyme radiation caries.All data collected to establish an Excel database, assure the result according to the original data checked, put the results into SPSS 16.0 statistical package for statistical analysis, use repetitive measure analysis of variance and rank sum test with test standard as bilateral a=0.05.Results1. The result of Resazurin disc method shows caries activity of nasopharyngeal carcinoma patients increased before and after radiotherapy (P<0.05).2. Dentin hypersensitivity increased after radiotherapy (P<0.05).3. BCA method to detect the total protein(μg/ml) in unstimulated whole saliva of NPC patients, the results were it was down (P<.05, content,875.04+114.93 before radiotherapy, content,773.71+116.46 after radiotherapy). SDS-PAGE shows molecular weight about 14.5 kD, 28kD,38 kD,56 kD,80 kD appear more stable. Among which the molecular weight about 14.5 kD,28kD,38 kD,56 kD were down after radiotherapy (P<0.05; but the protein of molecular weight about 80 kD was increased after radiotherapy (P<0.05)4. Immunoblotting to check the lysozyme and lactoferrin in NPC patients before and after radiotherapy, the results shows that the IOD and protein content percentage of lysozyme were decreased after radiotherapy (P<.05, IOD is 64.46+37.97 before radiotherapy,37.39+28.83 after radiotherapy; percentage of before and after radiotherapy is 6.80+9.84, 4.58+7.12 respectively);Iactoferrin has no significant difference(P>0.05 IOD of lactoferrin is 46.26+30.32 before radiotherapy,46.28+31.46 after radiotherapy; the content percentage is 2.50+1.62,2.82+1.62 respectively). The interaction between two proteins by analysis of variance showed that the two proteins have interaction (P<0.05). Conclusion:1. Both caries activity and dentin sensitivity had increased after radiotherapy.2. Four straps of molecular weight about 14.5kD,28 kD,38 kD,56 kD with IOD and protein content percentage decreased after radiotherapy, while 80 kD is higher after radiotherapy.3. The lysozyme IOD and the percentage of protein content in NPC patients is deceased after radiotherapy, but the lactoferrin IOD and the percentage of protein content in NPC patients has no significant change after radiotherapy...
