Effect Of Fengbaisan Decoction On The Expression Of MMP-9 And Its Inhibitor TIMP-1 In Brochus And Lung Tissue Of Rats With Chronic Obstructive Pulmonary Disease

Objective:To observe effects of Fengbaisan Decoction on chronic obstructive pulmonary disease (COPD) model rats morphology and the expression of matrix metalloproteinase-9 (MMP-9) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in brochus and lung tissue of rats with chronic obstructive pulmonary disease, and to study the therapeutic effects and the potential mechanisms of Fengbaisan Decoction.Methods:46 Wistar rats were randomly divided into two groups: the normal group and the model group. No intervention was applied in the normal group. COPD model was established by modified methods of combining fumigation and lipopolysaccharide(LPS) intra-tracheal dripping, and then rats of the model group were randomly divided into three groups:COPD model group, Ambroxol Hydrochloride treatment group and Fengbaisan treatment group. Model rats were treated respectively for successive 28 days, that is to say, the COPD model group were fed with normal saline daily, the Ambroxol Hydrochloride treatment group were fed with Ambroxol Hydrochloride, and the Fengbaisan treatment group were fed with the Fengbaisan Decoction. Observe the differencens on morphology in brochus and lung tissue by means of hematoxylin and eosin staining and iamge analyzer; Reverse transcription polymerase chain reaction (RT-PCR) was used to evaluate the mRNA levels of matrix metalloproteinase-9 (MMP-9) and tissue inhibitor ofmetalloproteinase-1 (TIMP-1); expressions of MMP-9 and TIMP-1 in brochus and lung tissue of rats were measured with immunohistochemical staining.Results:1. Histopathology change:in brochus and lung tissue of rats of COPD model group, Ambroxol Hydrochloride treatment group and Fengbaisan treatment group, the typical pathologic features of emphysema were displayed which were fit for the characteristic pathologic changes of COPD, and airway remodeling was obvious.We drew a conclution that the COPD model was established successfully. What s more, the pathogical changes of emphysema in lung tissue of rats of Ambroxol Hydrochloride treatment group and Fengbaisan treatment group were fewer than the COPD model group s.2. Results of Reverse transcription polymerase chain reaction (RT-PCR):Compared with the normal group, the expressions of MMP-9mRNA and TIMP-1mRNA in the COPD model group were increased significantly, there were significant differences (P<0.05); the expressions of MMP-9mRNA and TIMP-1mRNA in the Fenbaisan treatment group were decreased markedly as compared to the COPD model group, there were statistical differences (P<0.05).3. Results of Immunohistochemistry:Compared with the normal group, the expressions of MMP-9 and TIMP-1 in COPD model group were higher significantly, there were significant differences (P<0.05); the expressions of MMP-9 and TIMP-1 in the Fenbaisan treatment group were lower markedly as compared to the COPD model group, there were statistical differences (P<0.05).Conclusions:1.The typical pathologic features of chronic bronchitis and emphysema were displayed and rat COPD model was established successfully by exposed to cigarette smoke and injected lipopolysaccharide in trachea of rats.2. By measuring the thicknesses of airway wall and smooth muscle with image analyzer, the thicknesses of airway wall and smooth muscle were increased significantly in rats with COPD model,this inferred that airway remodeling was formed.3. By observing the expressions of MMP-9 and TIMP-1 in rats with COPD model, it witnessed that with overexpressions of MMP-9 and TIMP-1 in brochus and lung tissue, MMP-9/TIMP-1 imbalance was related with the pathogenesis of airway remodeling in COPD.This confirmed that proteinase-antiproteinase imbalance was one of mechanisms of COPD.4. Fengbaisan Decoction can reduce the expressions of MMP-9 and TIMP-1 in rats with COPD model, and adjust MMP-9/TIMP-1 imbalance to intervene in airway remodeling, those partly may be therapeutic mechanisms of Fengbaisan Decoction in chronic obstructive pulmonary disease.
