Objective To detect the expressions of Smo and Gli1 in induced primary hepatocellular carcinomas modes,study the relationship between Smo,Gli1 and the progress of HCC, explore the effect of VitD3 on Shh signaling pathway.Methods Totally 48 from 5 weeks to 6 weeks old health male SD rats were randomized into 3 groups, which were respectively implanted with nothing (Normal group),with DEN (Cancer group) ,with DEN and VitD3 (VitD3 group).The specimens were harvested separately at the end of 12 and 20 weeks.The expressions of Smo and Gli1 were detected by Western blot and Real time PCR.Results (1)In Normal group ,the expressions of Smo and Gli1 were not significantly different at the end of 12 and 20 weeks(P>0.05).In Cancer group or VitD3 group ,the expressions at the end of 12 weeks were higher than that at the end of 20 weeks(P<0.05).(2)At the end of 12 weeks or 20 weeks,the expressions of Smo and Gli1 in Cancer group were elevated compare with VitD3 group(P<0.05).The expressions of Normal group was lowest relatively(P<0.01).Conclusions Smo and Gli1 might participate in the progress of HCC.VitaminD3 may be an inhibitor of Smo and Gli1 in SHH signaling pathway. |
