| Normal thyroid cells independently take up radioiodine, iodine concentrations inside cells reach more than 20 to 40 times of that in blood,which makes it possible to treat thyroid carcinoma with 131I. During tumour progerssion,cellular de-differentiation is present,and usually accompanied by loss of iodide uptake ability.So,radiotherapy is less efficient. In this study, we observe the possiblity of ATRA and TSA alone or combined re-inducing its uptake in thyroid carcinoma in vitro and vivo,which demonstrates its potential for treating thyroid carcinoma.PARTâ… The Effect of ATRA and TSA alone or combined in Thyroid Carcinoma Cell LineObjective:To observe the influence of ATRA and TSA alone or combined on iodine uptake ability and cell proliferation in thyroid carcinoma cells.Methods: We detected the iodine uptake after induced by ATRA,TSA in different concentrations alone and combined in Thyroid Carcinoma Cell Line FTC-133. We detected the influence of ATRA and TSA alone or combined on the proliferation and morphology in vitro by MTT and HE. Results: The iodine uptake rate is respectively 23885±615.99cpm,13849±728.22cpm in group A低+T and A高+T. Differences between groups is statistically significant, F = 600.879, P = 0.000; OD is higer in drug combinations of low concentrations than that in other groups.Conclusion: Both ATRA and TSA can inhibit proliferation,induce cell differentiation, promote cell apoptosis in FTC-133. The idione uptake is involved by drug concentration; the inhibition of cell proliferation is concentration and time depended. The combination of ATRA and TSA can inhibit the proliferation in FTC-133,with less toxicity and stronger re-differentiation.PARTâ…¡The therapeutic effect of ATRA,TSAObjective:To study the iodine uptake ability and treatment effect in tumor-bearing nudes which were induced by RA,TSA..Methods:FTC-133 were subcutaneously injected into nude mice.After tumour formation, ATRA and TSA alone or combined was started to be administered to the tumor-bearing mice daily. At the 22st day,the tumor-bearing mice received the injection of 131I via enterocoelia. The biodistribution of 131I and imageing by SPECT were investigated. In the end of the experiment,histopathologic examinations of tumor samples were undertaken.Results:The imaging results showed that the tumor tissues accumulated the highest radioactivity in the ATRA combined TSA group at 12 h,while no in other groups.The difference of inhibiting tumour growth between the ATRA combine TSA group and control group is significant statistically(P<0.05).Conclusion:The iodine uptake of tumour tissue was enhanced in the tumor-bearing mice administered with ATRA combined TSA, which demonstrates its potential for treating Follicular thyroid carcinoma. |
