Study On The Extraction And Purification Process Of The Total Saponins Of Rubus Parviflolius L.

Several researches showed that TSRP has a variety of important pharmacological activities. Reported on the purification process of TSRP were very simple. Based on the references, this research had systematicly studied on the extraction and purification process of TSRP, which will provide the theoretical basises and technical supports for the further research or industrialization of TSRP .This research had systematicly studied on the ultraviolet-visible spectrophotometry for the content determination of TSRP; The reflux extraction process of TSRP by ethanol was optimized ; Studied on purification process of TSRP by D101 macroreticular resin. Major results of this research were:l. The method for the content determination of TSRP by ultraviolet-visible spectrophotometry has been established as: Under the control of oleanolic acid (OA), 546nm was been used for the maximum absorption wavelength of TSRP; The linear regression equation was established as A = 1.8059C-0.0063 ,R2 = 0.9971; Sample recovery was 98.15%, RSD= 3.89%; The results showed that the method can quickly and accurately determine the content of TSRP.2. The best extraction process was been established in the orthogonal which was based on single factor research in the research of the extraction process by ethanol solution for TSRP: The concentration of ethanol solution was 70%;Ratio to solid was 1:20;Extraction temperature was 80℃; Extraction time was 3h.3. In the research we had studied on the effects of a variety of macroreticular resin which were used for the purification of TSRP. The results showed that the D101 macroreticular resin was better for the purification of TSRP, which has higher desorption rate and resolution rate, and its adsorption rate has reached 23.96m /g, desorption rate has reached 83.78%; In the research of purification of TSRP by D101 macroreticular resin we had studied about effects of several factors to the adsorption and desorption. The result is:The sample (4.86mg/mL) adsorpted by 3BV/h; Desorpt the water soluble impurities with the 5BV of water first; And then eluted with 5BV of 50% ethanol solution and collect the desorption solution; Evaporated the desorption solution to dry for getting the crude sample of TSRP. Finally ,we found the purification of TSRP increasing ignificantly.
