Objective: To investigate the effects bone morphogenetic protein-2 (BMP-2),Thermo-chemotherapy (5-FU) and their combinition on the proliferation and apoptosis of human colon cancer cell(SW480) line, and to study the possible mechanism.Methods: We used BMP-2(25,50,100,200,400μg/L)to SW480 cells and detectded cell viability of SW480 cells by MTT assay. BMP-2(100μg/L) and 5-FU(50mg/L) were used to SW480 cells and their apoptosis rate was detected by flow cytometry (FCM). The protein expression of phosphatase and tensin homolog deleted on chromosome ten (PTEN) was detected by Western blotting. The mRNA expression of PTEN was detected by reverse transcription PCR (RT-PCR).Results: BMP-2 can inhibit the proliferation of SW480 cells .The apoptosis rate of sw480 cells treated Thermo-chemotherapy (5-FU)+BMP-2 was significantly higher than those of BMP-2 treated and those of Thermo-chemotherapy(5-FU) treated cells(P<0.05 ).BMP-2 can improve the expression of PTEN protein. The expression of PTEN mRNA have no difference in groups.Conclusion: BMP-2 can improve the sensitivity of Thermo-chemotherapy.BMP-2 combined with Thermo-chemotherapy (5-FU) treated can induce apoptosis of SW480 cell line, and the mechanism may be through increasing the protein expression of PTEN.
