| Objective It has been reported that the abnormal expression of zinc ions were related with the genesis and development of epilepsy. Exploring the changes of the related genes may be benefical to enhance the mechanisms of epilepsy. The study aimed to detect the expression genes related with zinc-binding at mRNA levels in the patients with drug-resistant epilepsy and explore the relation between zinc and refractory epilepsy.Methods The temporal samples from forty-eight patients with drug-resistant epilepsy were enrolled for this study. At first, we used large-throughput microarray scanning, and compared with the control, and selected the genes related with zinc ion, then applied RT-PCR testing.Results Compared with the control, the results of microarray scanning showed that the expression of the four genes related with zinc-binding TRAF3,RYBP,CNOT4 were higher in the refractory epilepsy, and RT-PCR results also showed that the expression of TRAF3, RYBP, CNOT4, NR4A2 mRNA were significantly higher in the patients with drug-resistant epilepsy than those in the control, which were identical with the gene microarray(P<0.05).Conclusion Increased expression of TRAF3, RYBP, CNOT4, NR4A2 related with zinc-binding in the temporal tissues may be associated with human drug-resistant epilepsy, and zinc seems to play a role in the pathogenesis of drug-resistant epilepsy. |
