Chito-oligosaccharides is the product after depolymerization of chitosan. The molecular structure of monomer of chitosan before poly identical unsolved. With the degradation of chitosan, its degree of polymerization decreased, the molecular weight decreases, water-soluble gradually increased, physical activity is relatively increased. Therefore, the large increase and broaden its scope of application, there are a lot of advantages compared with chitosan.This study focuses on the preparation of chitin and chitosan optimal conditions, and by the hydrolysis process and membrane separation technology to prepare a molecular weight of 300DaB>D>C, the alkali concentration> temperature> liquid ratio> time. Under the above conditions in the production of chitin ash content≤2%, crude protein content of≤7%; chitosan was dried products was 20.4%, average degree of deacetylation was 84.8%.②by crustaceans and DEAE-chitosan oligosaccharide removal (O2-·)and (·OH) of the experiment, found that concentration of Chitooligosaccharides 10 mg / mL, the for (O2-·)and (·OH) removal rate can reach 60%; concentration of 35 mg / mL, the for (O2-·)and (·OH) scavenging rate can reach 90%. DEAE-chitosan concentration of 10 mg / mL, the for (O2-·) and (·OH) removal rate reached 21.3% and 24.9%; concentration of 35 mg / mL, the for (O2-·)and (·OH), clearance rates were 31.92% and 36.92% achieved. Oligosaccharides with crustaceans and DEAE-chitosan in mice fed 30 d, the determination of SOD, T-AOC, CAT activity and MDA content indicators, the experimental results: COS medium dose group (400 mg/kg), COS High dose group (800 mg/kg) and D-COS high dose (800 mg/kg) in mice serum and liver SOD, T-AOC, CAT activity was significantly higher, MDA levels were significantly lower than control group.③Chitooligosaccharides containing medium and DEAE-chitosan-old flies were fed 30 d, respectively, at 0 d, the first 10 d and 20 d measured SOD and CAT enzyme activity, MDA content and the fruit fly life; Results: In the first 10 d and 20 d, COS middle dose group (0.8%), COS high dose group (3.2%) and D-COS high dose group (3.2%) of the Drosophila tissue SOD, CAT significantly higher activity, MDA content was significantly lower than the control group was significantly higher in drosophila.④the scope of the crustacean different molecular weight oligosaccharides and DEAE-chitosan adsorption experiments in vitro and animal experiments, the results show that: 300Da0.05) , COS-L mice spleen index reached 7.56±0.42 mg/g, significant effect (P<0.05), COS-M, D-COS-H, and COS-H group of the spleen index of 8.98±0.68, respectively , 9.06±0.78 and 10.02±0.37 mg/g, effect was significant (P<0.01); D-COS-L mice thymus index reached 4.01±0.34 mg/g, significant effect (P<0.05), COS-L, COS-M, D-COS-H, and COS-H thymus index of mice, respectively, 4.55±0.21,5.18±0.22,5.98±0.29 and 6.42±0.27mg/g, is extremely significant (P<0.01) .
