Objective The study was performed to dynamicly view the changes of liver function and the impairment of hepatic vascular endothelial cells of early sepsis rats,and to briefly discuss their correlation and possible mechanism.The sepsis model was induced by cecal ligation and puncture.Method :66 SD rats,male and female informal,were randomly divided into three groups as follows,(1)normal control group(n=6),(2)sham operation group(n=30),(3)sepsis group(n=30). Sham operation group and sepsis group were further divided respectively into 3,6,12,24 and 48h subgroups after CLP,each subgroup of sham operation and sepsis group had six rats.All rats were measured changes of ornithine carbamoyltransferase (OCT) ,sE-selectin,TNF-αin blood serum,observed impairment of vascular endothelial cell using electron microscope.Calculated liver wet weight/dry weight ratio(W/D), after weighing wet weight and dry weight of liver.Results:1.The activity of OCT was higher at 3 hours after CLP(5.9805±0.07u/l),significantly more than normal and sham operation group(P<0.05), and gradually rised with time-lapse. 2.The serum levels of TNF-αin sepsis group after CLP was significantly higher than normal and sham operation group(P<0.01),reached maxlmum(390.94±27.70pg/ml) at 24 hours point.Then,there was a downward trend,but still higher than the normal and sham operation group(P<0.01). 3.The serum level of sE-selectin began to increase at 3 hours after CLP(1.6292±0.01ng/ml),significantly higher than normal and sham operation group(P<0.05), and gradually rised with time-lapse. 4.The hepatic water content were higher than the normal and sham operation groups(P>0.05),and significantly higher than those at 12 hours after CLP (P<0.05). 5.The damage of vascular endothelial cells,and the score of vascular endothelial cell gradually rised with time-lapse. 6. sE-selectin was positive correlated with OCT and TNF-α, Spearman coefficient correlation were 0.947 and 0.781(P<0.01).Conclusions 1.The damage of endotheliocyte was gradually aggravated with the development of sepsis, endotheliocyte's impairment may play an important role in sepsis. 2. sE - selectin participate the pathogenesis and development of sepsis,TNF–αis a key cytokine in inducing upregulation expression of sE– selectin. 3.The damages of VEC and liver function had obviously direct correlation,which hints endothelial cell may play an important role in acute hepatic injury...
