| Background Psoriasis is a common chronic inflammatory skin disorder with strong genetic component and multiple environment factors. We found a distinguished correlation of rs3751385 polymorphism in GJB2 gene cluster with psoriasis vulgaris in Chinese Han Population by psoriasis genome-wide association study(GWAS). .This study investigated the relationship between GJB2 and subphenotypes of psoriasis.Objectives Based on stratifying analysis of the age of onset, family history and clinical types, the study compared the genotype distribution of rs37513851 polymorphism within GJB2 gene with some clinical features of psoriasis vulgaris in the Han Chinese population to lay solid foundation for the pathogenesis of psoriasis.Methods We analyzed the distributions of genotype and allele of rs3751385 polymorphism within GJB2 gene in 6,693 cases and 1,0718 normal controls using the Human 610-Quad BeadChips, which derived from the psoriasis genome-wide association study(GWAS).The appropriate application of social science statistical software package SPSS 10.0 was used for statistical analysis of data. .Results 1.The distribution of alleles and genotypes for rs3751385 were significant differences between cases and controls(P=3.50×10-9, 1.90×10-8, respectively).Then cases were stratified from onset, family history and lesion type, there were also significant differences between clinical subgroups and controls in distribution of allele and genotype(all P<0.05).2. The distribution genotypes for rs3751385 showed significant associations between family cases and sporadic cases (x2 = 8.006, P = 0.018),but there weren't significant differences between c family cases and sporadic cases in distribution of allele(x2=3.121, P=0.077).3.There weren't significant differences between the pediatric onset psoriasis patients and the adult onset psoriasis patients in distribution of allele and genotype of rs3751385 (x2=1.835, P=0.176;x2=1.894, P=0.388).4. The difference in frequencies distribution of allele and genotype for rs3751385 between the guttate and plaque cases did not reach the statistical significance(x2=0.135, P=0.713 ;x2=0.14,P=0.932).Conclusion 1 1. The SNP rs3751385 in GJB2 gene is associated with the susceptibility of psoriasis vulgaris in the Chinese Han population. 2. The SNP rs3751385 in GJB2 gene is associated with family history of psoriasis vulgaris 3. The SNP rs3751385 in GJB2 gene isn't associated with age of onset and clinical type of psoriasis vulgaris. |
