Cerebral infarction is also called ischemic stroke (cerebral isehemic stroke, CIS), which manifests itself in corresponding neurological deficits according to the ischemic regions, anoxic zones or necrotic areas caused by the disorder of blood circulation.According to the pathogenesis and the clinical manifestations, we can divide cerebral infarction into thrombosis,embolism and lacunar infarction. The clinical manifestations are varied closely related to the infarct location and the collateral circulation.Cerebral infarction is the most common type of cerebrovascular disease, accounting for about 75%, which has high morbidity,high mortality, and easily recurs.The recurrence of stroke is more dangerous.Therefor,early detection,early diagnosis and early treatment play an crucial role for improving the prognosis of patients with infarction.Currently multiple mechanisms are known to participate in the pathophysiological process of cerebral infarction,including inflammatory mediators,apoptosis,oxidative stress and so on.Inflammatory plays a key role in the pathophysiology of acute cerebral infarction.Infarction might cause massive release of inflammatory mediators,which results in serious damage to normal brain tissue. Growing evidence shows that excessive release of inflammatory factors is closely related to the occurrence,the development and the severity of cerebral infarction, maybe as an important trigger for cerebral infarction. Studies have shown that inflammatory factors (such as NF-a, IL-6, IL-3, IL- 8, IL-26 and IL-10)increased not only in immune cells, but also in neurons and glial cells in patients with acute cerebral infarction.Pentraxin is a calcium-binding protein family with immune defense properties ,characterized by a cyclic pentamer composed of five identical subunits with non-covalent bond. Currently the family members studied most are C-reactive protein (CRP) and serum amyloid A (SAA) and other short-chain pentamer protein. Pentraxin 3 (PTX3) is a member of the family, as a long-chain pentamer protein, belonging to acute phase proteins. Human PTX3 gene is located on chromosome 3 ,q5 region. Current research on PTX3 focuses on acute coronary syndrome, hypertension and other diseases.Zhaofa He indicated that PTX3 plays an important role for monitoring the occurrence and progression of hypertension. In inflammatory response, PTX3 appears later than hs-CRP, but it lasts longer, contributing to its being a marker for persistent inflammation. In conclusion, PTX3, as the main damage product from the heart and blood vessels during acute phase reaction, is more likely to became a sensitive and specific biological indicators than hs-CRP from liver. For PTX3,we have gotten some preliminary understanding on the cell source, biological activity, molecular structure, the relationship with some diseases of, but PTX3 is a newly discovered inflammatory markers.The relationship between PTX3 and cerebral infarction and the role in the development of cerebral infarction require further study.Objective:Measure PTX3 level in plasma of patients with cerebral infarction and normal controls by enzyme-linked assay (ELISA) to study the correlation between PTX3 and cerebral infarction, providing the basis for the diagnosis of diseasesMethods:1 Subjects: We choose 37 acute cerebral infarction patients in Second Hospital of Hebei Medical University from October 2010, including 27 males and 10 females, aged from 42 to 75 , average 61 years old. All patients meet the diagnostic criteria revised by the Fourth National Conference in Chengdu in 1995 and are confirmed by brain MRI or CT. Subjects can't have immune system disease, infectious diseases or malignancies ,and can't take drugs that affect immune function. Choose 21 healthy patient over the same period as the normal controls,17 males and 4 females, aged from 37 to 75, mean 60 years old. There is no difference between sex and age.2 Specimen collection: Pump 2 ml fasting venous blood in patients with acute cerebral infarction early the next morning and in normal controls at 8 am; put into EDTA-tubes;centrifuge(3000r/min) 20 minutes; separate plasma;store at - 80℃until test.3 Plasma PTX3 determination: PTX3 test kit is bought from Shanghai Blue Gene Biotechnology Co, Ltd.and operated according to reagent instructions by enzyme-linked immunosorbent assay (ELISA).4 Statistical analysis Proces the data using statistical software SPSS13.0 ; the obtained data with SPSS statistical software processing, testing results to mean + standard deviation , said the x + s usingTo P <0.05 indicates significant difference. Results:Plasma PTX3 level in patients with cerebral infarction is higher than that of normal control group .The differences are statistically significant (P < 0.05).Conclusion: Measure PTX3 level in cerebral infarction patients'plasma and normal controls by enzyme-linked assay (ELISA) .Plasma PTX3 level in cerebral infarction patients is higher .The differences are statistically significant (P < 0.05).It indicates that PTX3 is closely related to the occurrence of cerebral infarction as an acute inflammatory response protein. PTX3 increases after cerebral infarction,causing massive release of inflammatory mediators ,which leads to serious central nervous system injury. The inflammatory response in cerebral infarction patients may relate to increased PTX3 concentration. Analysis of the relationship between PYX3 and cerebral infarction occurrence and development will play an important role in the diagnosis and treatment of acute cerebral infarction.
