Objective: To investigate the expression of connective tissue growth factor(CTGF) and smooth muscle alpha-actin (α-SMA) in myocardium of streptozotocin-induced diabetic rats, and to further explore the protective effects of Didangqigui Recipe on vascular endothelium in STZ-induced diabetic rats. and to explore its mechanism of action on prevention and treatment of diabetic cardiomyopathy,and provide experimental evidence for its clinical applications.Methods: 80 SD rats were selected, 10 rats were set randomly as normal group, and others were injected STZ(60 mg·kg-1) to establish the diabetic model. After model was established, and 55 model rats were randomly diveded into five groups: the diabetic model group, the gliclazide group, Didangqigui Recipe Conventional-dose group, Didangqigui Recipe high dose group, Didangqigui Recipe and gliclazide group,and they were treated respectively. With treatment being given respectively, myocardial tissue in each group were collected after treatment. The expression of CTGF andα-SMA was detected by immunohisto- chemical staining. The expression ofα-SMA mRNA was detected by RT-PCR and real time PCR . Through the light and electron microscope rat heart tissue structure change.Results: Compared with the diabetic model group,The intervention group can impr-ove STZ model of rats which were much more food,drink,urine,in general,and can be inhibited weight loss of the rats, Didangqigui Recipe Conventional-dose and Didangqigui Recipe and gliclazide groups which improved significantly (P<0.05).Compared with the diabetic model group significant decreasements of the fasting blood glucose and glycated hemoglobin were observed especiallly in gliclazide and Didangqigui Recipe and gliclazide groups (P<0.05). Expression of CTGF,α-SMA andα-SMA mRNA in myocardial tissue in rats dramatically decreased in Didangqigui Recipe Conventional-dose and Didangqigui Recipe and gliclazide groups(P<0.05). The intervention group had improved myocardial structure, especiallly in gliclazide and Didangqigui Recipe and gliclazide groups.Conclusion: Through this experimental study,it is impossible that Didangqigui Recipe have a certain therapeutic effect on STZ-induced diabetic cardiomyopathy rats.The mechanism was probably by adjusting the glucose metabolism disorder, and abnormal expression of CTGF,α-SMAandα-SMA mRNA in myocardial tissue in rats is another of the important mechanism that leads to diabetic cardiomyopathy of rats,and improve myocardial systolic and diastolic function, and delay the occurrence and development of diabetic angiopathy.
