Fructus schisandrae, commonly known as Mountain prickly ash, Cheng Tuozi, Medicine Fructus schisandrae, Surface cane and so on. Fructus schisandrae belong to Magnoliaceae, which is perennial deciduous plants not need to be repeated planting. It also has the ornamental value. Fructus schisandrae generally have sweet, sour, piquancy, bitter, salty. However, Fructus schisandrae is classified into the north and south. It has the actions of nourishing generate fluid, tonifying kidney and nourishing heart, absorbing, consolidating and astringent. Lignin is the most important effective constituent in the Fructus schisandrae, it also contains volatile oil, polysaccharide and triterpenic acid. Lignin compound have the actions of protect liver, decrease enzyme and protect Cardiovascular. Modern pharmacology study suggests:Lignin mainly contains Schizandrin A, Schizandrin B, Schisandrin in the Fructus Schisandrae. It can obviously reduce the levels of serum Alanine aminotransferase, promote generating liver glycogen, protecting Hepatocyte for patients who are Hepatitis.There are abundant Fructus Schisandrae resources in the northern China, is the main raw materials to extract Lignin. But the current condition is low extraction rate, low purification degree, and this seriously influences the Lignin's utilization prospects. In order to develop and utilize Fructus Schisandrae resources preferably, this research in order to improve the extraction rate for the purpose, take the Schizandrin B, schisandrin as research objects, utilize ultrasonic cell disruptor, Macroporous resin, HPLC to extraction, isolation and purification, detection. On this basis, conduct Bacterial restrain experiment using Schizandrin B and Schisandrin.1,By single factor experiment and the orthogonal experiment, we got the best extraction conditions, it is:ultrasonic power 600W, solid-to-liquid ratio 1:40, ultrasonic time 15min, solvent strength 75%.Through the best extraction conditions got Schizandrin B and Schisandrin. The former extraction dosage is 28.90mg, extraction ratio is 5.78%, the latter extraction dosage is 30.74mg, extraction ratio is 6.15%.2. Four different models of Macroporous resin were used to purify Schisandrin and Schizandrin B, results show that AB-8 is the best resin.Study the Schisandrin and Schizandrin B's static adsorption dynamics test through the Macroporous resin, we obtained at 4h achieved stable equilibrium.Study the Schisandrin's dynamic adsorption dynamics test through the macroporous resin, we got the best conditions:feed concentration is 0.07mg/mL, flow rate is 1.5mL/min, Elution velocity is 0.75mL/min, eluant dosage is 80mL. Study the Schizandrin B's dynamic adsorption dynamics test through the Macroporous resin, we got the best conditions:feed concentration is 0.05mg/mL, flow rate is 1mL/min, elution velocity is 0.5mL/min, so we confirmed that the eluant dosage is 70mL.3. By means of Schizandrin and Schizandrin B on Escherichia coli, Aspergillus niger, beer yeast of bacteriostatic experiment, confirm that the best bacteriostatic efficacy.Through the experiment of minimal inhibitory concentration, we can determine Schizandrin inhibit Escherichia coli's minimum inhibitory concentration between 0.0625g/mL to 0.125g/mL; minimum inhibitory concentration of Aspergillus niger between 0.125g/mL to 0.25g/mL; minimum inhibitory concentration of beer yeast between 0.03125g/mL to 0.0625g/mL.Through the experiment of minimal inhibitory concentration, we can determine Schizandrin B inhibit Escherichia coli's minimum inhibitory concentration between 0.0625g/mL to 0.125g/mL; minimum inhibitory concentration of Aspergillus niger between 0.125g/mL to 0.25g/mL; minimum inhibitory concentration of beer yeast between 0.03125g/mL to 0.0625g/mL.
