Partâ… EFFET OF IFENPRODIL ON SURVIVIN EXPRESSION AFTER CEREBRAL ISCHEMIA REPERFUSIONObjective:To investigate the expression discipline of survivin after perifocal area of cerebral after ischemia-reperfusion (I/R) in rats, and to identify the effect of ifenprodil on Survivin after I/R. Method:Seventy-two Wistar rats were randomly divided into three groups:shame group, model group and ifenprodil group. The middle cerebral artery occlusion reperfusion (MCAO/R) model was established by thread ligation. After operation in the shame group,2 hours ischemia of middle cerebral artery following 3 hours,24 hours,72 hours and 7days reperfusion in model group and in ifenprodil group. Rats were sacrificed and the expression of Survivin in hippocampus of ischemia side was detected by immunofluorescence. Result:fluorescence intensity of the shame group was very weak, just showing the basal expression; while fluorescence intensity of model group and ifenprodil group have almost the same dynamic trend, it raised 3 hours after I/R, increased continuously from that time and reached the peak at 72 hours, and then decreased afterward, P<0.05 as compared with the shame group; fluorescence intensity of ifenprodil group were stronger than model group at the same time point(P<0.05). Conclusion:1. Expression of Survivin increased after MCAO/R, and reach the maximum at 72 hours; 2. ifenprodil has a protective effect on damage of I/R.Part II EFFET OF IFENPRODIL ON MUSASHI1 EXPRESSION AFTER CEREBRAL ISCHEMIA REPERFUSIONObjective:To investigate the protective effect of ifenprodil on model of ischemia-reperfusion (I/R) in rats and to identify expression discipline of Musashil. Methods:Eighty-four Wistar rats were randomly divided into shame group, model group and ifenprodil group, each twenty-eight. The right middle cerebral artery occlusion (MCAO/R) model was established by thread ligation. After operation in shame group,2days,3 days,7 days and 14days reperfusion after 2 hours of ischemia of middle cerebral artery in model group and ifenprodil group.7 Rats were sacrificed for sample collection,1 of them was for electron microscope to observe the ultrastructure,the left were used for detection of Nissl's staining and immunohistochemisty staining. Results:â‘ Compare to the shame group, result of electron microscope in model group:chromatin gathered marginly, pericaryon and organelle in it swelled markedly, optical density of cytolymph decreased; Nissl's staining:number of survival neurons dereased, cell body swelled, nissl's body dereased or disappeared; staining of immunohistochemisty:Musashi1 positive reaction cells raised 2 days after reperfusion, increased sharply 3days and keep at a high level at until 14 days.â‘¡Compare to the model group, result in ifenprodil group, electron microscope indicate:pericaryon and organelle in it swelled slightly; Nissl's staining:number of survival neurons and nissl's body increased; immunohistochemisty staining:Musashi 1 positive reaction cells had the same expression trend with model group, but optical density value was higher than that in model group. Conclusion: ifenprodil can promote the expression of the nerve regeneration factor Musashi1 after I/R, has a positive effect on damage of I/R.
