Objective:The expression pattern and correlation of hypoxia-inducible factor-la and stromal cell derived factor-1 in the ischemic retinal diseases and retinal ischemia-reperfusion.Methods:56 adult SD rats with normal eyes were randomly divided into surgery group and sham surgery group, all the two groups were operated at 0,6th,12th,24th,48th,72th,144th hour.6 rats and 2 rats were operated in the operation group and sham surgery group at the same time respectively. We applied method of carotid artery ligation to establish animal models of retinal ischemia and reperfusion.60 minutes later, ligation surgery group restored blood flow perfusion. After that, we removed the eyes at 0,6th,12th,24th,48th,72th,144th hour, cut off the cornea, removed the lens, clear vitreous, washed retinal specimens repeatedly with fresh water slowly. Alcohol dehydration and paraffin-embedded retinal specimens were completed after all above, methods of immunohistochemical and average optical density were applied in each group at different phases in order to detect positive expression of HIF-la and SDF-1.Result:HIF-la and SDF-1 was not expressed or slightly expressed in the sham group of retinal. HIF-la and SDF-1 expression at the beginning 0h,6h expression increased in retinal operation group,12h strongest expression,24h expression was decreased, after the expression was gradually decreased over time, animal model group compared with the sham group difference was statistically significant (P<0.01).HIF-1a and SDF-1 expression in consistent with the positive correlation.Conclusion:Hypoxia-inducible factor-1a (HIF-1a) and stromal cell derived factor-1(SDF-1) expression closely relate to retinal ischemia-reperfusion injury, while a collaborative relationship between the two.
