| Objective:To investigate the anti-influenza effects of EC on immune function in mice loaded with restraint stress.Methods:(1) The changes of survival rate of mice was investigated. The changes of spleen, lung and thymus index, the total number of spleen in restraint stressed mice after restraint stress were observed and compared.(2) In order to study the anti-influenza effects of EC on immune system in restraint stressed mice, the index of immune organs, the total number of spleen and thymus lymphocytes, the subsets of lymphocytes Th(CD3+CD4+) and Ts(CD3+CD8+) in spleen was investigated. The spleen NK cell activity was observed after treated with BEC. KM mice were randomly divided into 5 groups with 5 mice each group:normal control group, virus group, restraint and virus group, EC-L group and EC-H group. All mice were restraint for 18 h except normal group. All mice were sacrificed after treated with EC for 7 days.the expression of NP, MP, INF-γ, IL-12, IL-18 mRNAs in lung were observed.Results:(1) The results shows that the survival rate of model group was decreasing compared with virus group.The results showed that the spleen index, the total number of spleen lymphocytes obviously decreased, lung index significantly increased.(2) EC-L and EC-H could significantly decrease the index of lung,the total number of spleen index and spleen lymphocytes in restraint stressed mice increase. EC-L and EC-H could obviously elevate the percentage of Th(CD3+CD4+) and reduce the percentage of Ts(CD3+CD8+) in spleen lymphocytes of stressed mice, resulting in the increase of Th/Ts ratios which indicated the recovery of immune function. It is showed EC significantly elevated spleen index and the total number of spleen lymphocytes. RT-PCR analysis showed EC markedly up-regulate the expression of INF-γ, IL-12, IL-18 mRNA, down-regulate the virus NP, MP expression of stressed mice in the lung.Conclusion:EC had obvious anti-influenza effects and preserved the function of immue system... |
