Bleeding is one of the causes of death in the injured patients, rapid and effective control of bleeding can significantly reduce the mortality rate. Surgery is one of the rapid and effective methods of controlling bleeding, but surgery can not fully control the bleeding, the hemostatic material can greatly reduce the amount of bleeding. Currently there are a lot of hemostatic material,some hemostatic inaccurate, some expensive, some inconvenient to use, etc. We have developed chitosan hemostatic powder, through the formulation of reform and structural change, and adding some calcium, zinc. The chitosan hemostatic powder has many advantages, bleeding, eliminating inflammation and promoting wound healing, anti-adhesion, cheap, easy to use, etc. we would prove whether the chitosan hemostatic powder has the above-mentioned functions and provide the basis for clinical applications through animal experimentation.Part one EXPERIMENTAL STUDY ON HEMOSTASIS OF A NEW CHITOSAN HEMOSTATIC POWDERObjective To study hemostasis of a new chitosan hemostatic powder.Methods Twenty-four adultSD rats were made the models of l iver injury, male or female, and weighing 210-240 g. They were divided into three groups randomly (n=8) depending on different hemostatic powders. The incision of the l iver was treated with 300 mg Yunnan baiyao(group A1), chitosan hemostatic powder of pH6.5 (group B1) and pH7.5 (group C1), respectively. The bleeding time and bleeding amount were recorded. In vitro, with the modified Ree-White method, 2 mL artery blood from New Zealand white rabbit was added into the 0.2 mL solution of Yunnan baiyao, chitosan hemostatic powder of pH6.5 and pH7.5 (concentration of 0.2 mg/mL), respectively. The blood coagulation time was recorded. The chitosan blood clots of group B2 and group C2 were observed with scanning electron microscopy (SEM).Results The bleeding time of group A1, group B1 and group C1 was (292±31), (261±23), and (224±28) s, respectively, the bleeding amount was (1.63±0.21), (1.47±0.18), and (1.18±0.17) g, respectively, showing statistically significant differences between groups B1, C1, and group A1 (P < 0.05), between group C1 and group B1 (P < 0.05). The blood clotting time of group A2, group B2, and group C2 was (653±41), (255±20),and (202±11) s, respectively, showing statistically significant differences between groups B2, C2, and group A2 (P < 0.05),between group C2 and group B2 (P < 0.05). The SEM showed that the blood cells of group B2 and group C2 gathered aroundthe chitosan. Conclusion Chitosan hemostatic powder of pH7.5 has good hemostasis.Part two EXPERIMENTAL STUDY ON HEMOSTASIS AND TISSUE COMPATIBILITY OF CHITOSAN HEMOSTATIC POWDER AND STARCHY HEMOSTATIC POWDERObjective To study hemostasis of hemostatic powder and starchy hemostatic powder.Methods Forty-eight adultSD rats were made the models of l iver injury and the models of femoral artery injury, male or female, and weighing190g-210g. They were divided into three groups randomly (n=8) depending on different hemostatic powders. The incision of the liver was treated with 200 mg chitosan hemostatic powder (group A1), starchy hemostatic powder (group B1)and control group (group C1), respectively. The bleeding time and bleeding amount were recorded. The incision of the femoral artery was treated with 500 mg chitosan hemostatic powder (group A1), starchy hemostatic powder (group B1)and control group (group C1), respectively. The bleeding time and bleeding amount were recorded.After two weeks, the incision of the liver of group A1 and group B1 were observed pathological and observed with transmission scanning electron microscopy (TEM).Results The group A1 and group B1 can stop bleeding within 1.5 minute, the bleeding amount was (0.63±0.08),and(0.90±0.11), respectively. but the group C1 cannot stop bleeding within 1.5 minute. Showing statistically significant differences between groups A1 and group B1 (P < 0.05). The group A2 and group B2 can stop bleeding within 1.5 minute and 2.5 minute, respectively, the bleeding amount was (2.08±0.35),and(2.48±0.30), respectively. but the group C2 cannot stop bleeding within 2.5 minute.Showing statistically significant differences between groups A2 and group B2 (P < 0.05). The microscope showed more new liver cells and less Inflammation of group A1 were observed,the TEM showed that the liver cell nucleus and organelles of group A1 were more integrity.Conclusion Chitosan hemostatic powder has good hemostasis and tissue compatibility.
