Mulberry is deciduous shrub or arbor, all parts of Morus can be used as medicine in traditional medicine, All dynasties of Chinese Pharmacopoeias have a record of medicinal efficacy of leaves,mulberry branch,cortex mori and sorosis.Mulberry branch contain bioactive substances,such as flavonoids,alkaloids,sterols,coumarin, stilbene and terpenoids compounds etc. Which flavonoids have wild biological activity,such as antioxidant,anti-mutation,anti-aging and anti-tumor etc. But it has not been effectively utilized. Mulberry branch as raw material, extraction purification and antioxidant activity of flavonoids from mulberry branch are systematically studied in this paper,The main contents and the results are as follows:1. Extract technology research of total flavonoids from mulberry branchThis study investigated extraction time, ethanol concentration, raw material mass concentration, and temperature on extraction effect of total flavonoids. On this basis, according to orthoganol test, range and variance analysis,It was determined that the optimal extraction condition was as follows:extraction temperature 80℃, mass concentration of liquid materials 33g/L, ethanol concentration 70%, and extraction time 120min. Under these conditions, the extraction rate could reach 1.01%.2. Isolation and purification of total flavonoids from mulberry branchThis study investigated the purification of mublbery total flavonoids polyamide of the static and dynamic adsorption technical parameters. The sample volume of static adsorption was 1:274 g/ g (pure flavonoids volume:resin volume), the sample volume of dynamic adsorption was 1:61 g/g. the sample Concentration was 2.149mg/mL, the adsorption and resolution velocity both was 1mL/min, adsorption time was 1h and the eluent was 70% ethanol, Under these conditions,the elution of flavonoids could be up to 92.29%.3. Purification and identification of morin from mulberry branch95% ethanol extraction, ethyl acetate extraction and polyamide resin column chromatographic purification is use to purify morin in this paper. The results show that this method can obtain high purity morin.Thin layer chromatography show that using chloroform:methanol:acetic acid (15:5:2) as developing solvent and 1% aluminum nitrate as color reagant, we could observe green fluorescence of morin at365nm UV lamp. For qualitative analysis by High performance liquid chromatography showed that, using the symmetry (?) C-18 (4.6 x 150mm) column,methanol:water: phosphoric acid (50:49.8:0.2) as mobile phase, flow rate was 1mL/min, column temperature was 30℃, Under these conditions, morin has excellent chromatographic peaks at a wavelength of 370nm.4. Antioxidant activity in vitro reserchUsing vitimin E as control, evaluate the antioxidant capacity of rutin, quercetin, morin and total flavonoids from mulberry branch From two aspects of the free radical scavenging and reducing power in this paper.The results showed that the scavenging of DPPH·, quercetin had the strongest scavenging ability,was 1.4 times as power as vitamin E, morin and total flavonoids scavenging capacity was similar,but both were weaker than quercetin, rutin scavenging capacity was the weakest. In 0.02-0.08mg/ml range, quercetin had the strongest reducing power, morin and total flavonoid reducing power was similar to vitamin E,but both were weaker than quercetin,rutin had the weakest reducing.5. Comparison of total flavonoids content from different varieties of mulberry branchTotal flavonoids of several widely grown cultivars mulberry branch were determined in this paper, which material from the garden of mulberry germplasm resources of southwest university.the result showed that different varieties of mulberry have different flavonoids content,the order as follows:Jialing 20> Xinong6071> Guiyou62> Jialing 16> Husang 32> Yunsang 1> 7920.
