| ã€Objective】To investigate the relationship between the expressions of lymphoma/leukemia-2 (bcl-2)and the breast cancer resistance protein (BCRP) with clinical general parameters andchemotherapy efficacy in untreated diffuse large B cell lymphoma(DLBCL)。.ã€Methods】(1)A group of 40 previously non-treated patients with DLBCL qualified for these studies.Clinical advancement of the disease was classified according to the Ann Arbor scale. Accordingto the expression of bcl-2 and BCRP,all the patients were classified to several groups. All thepatients were subjected to 6-8 times the standard CHOP protocols of chemotherapy andevaluated response to chemotherapy.For all patients were followed up for six months.(2)The expression of bcl-2 was detected by way of immunohistochemical LASB method, ifthe percent is above 25%,we think the cells are positive.(3)Using semi-quantitative reverse transcription polymerase chain reaction ( RT-PCR) assayandβ-actin as positive reference ,we detected t he expressions of BCRP gene in 40 DLBCLsamples. The expression of BCRP mRNA in MCF-7/ADR as the positive control .(4)χ~2 analysis was used to study the correlation of the expression of bcl-2 and BCRP inDLBCL cells with general clinical parameters and chemotherapy efficacy.ã€Results】(1)We did not find any statistical significant correlation between expression of bcl-2, BCRPand clinico-pathological data (age, sex) P>0.05,but there was significant statistical correlationbetween expression of Ann Arbor stage and chemotherapy efficacy for BCRP( P<0.05).(2)The positive expression rate of bcl-2 and BCRP protein were 60% (24/40) and 37.5%(15/40) in 40 patients with DLBCL respectively. There was significant differences betweenpositive group and negative groups(χ~2= 5.7618,P<0.05;χ~2= 6.5541,P<0.05)ï¼›(3)There was no significant correlation between bcl-2 and BCRP gene in effective treatmentgroupre and therapies group or progress group(χ~2=2.0263,P>0.05).ã€Conclusion】(1) BCRP may be in DLBCL primary much medicine drug-resistant plays an important role,thus it may be useful prediction of chemotherapeutic treatment and risk of relapse.(2) There was no significant correlation between bcl-2 and BCRP gene in positive group andnegative groups.But united detection of bcl-2 and BCRP gene play an important role in evaluating DLBCL outcome. |
