Objective:The purpose of this topic is studying protective effect of adenosine and adenosine with lidocaine pretreatment on serum SOD, MDA and myocardial infarct size, compared the protection of myocardial ischemia-reperfusion between them.The purpose is to guide a clinical application.Methods:I/R model in vivo rats were established. Healthy male SD rats (n=32) were divided into 4 groups(8rats in each group):sham operation group (C group):open the chest and braid without deligation the blood vessel, the anesthesia maintain 150min; ischemia-reperfusion group (I/R group):left anterior descending (LAD) occlusion for 30min and reperfusion for 120min, without any intervention; adenosine group (AD group):left anterior descending (LAD) occlusion for 30min and reperfusion for 120min, the group was given adenosine 305μg/kg·min for 30min through the femoral vein before reperfusion; adenosine and lidocaine group (AL group):left anterior descending (LAD) occlusion for 30min and reperfusion for 120min, the group was given adenosine 305μg/kg·min and lidocaine 608μg/kg·min for 5 min through the femoral vein before reperfusion. TTC method was used to measure myocardial infarction area; Thiobarbituric acid method was used to observe the concentration of MDA; Xanthine oxidase method was used to observe SOD activity of each group.Result:1. The results of myocardial infarct size:Compared with sham-operated group rats (0), myocardial infarct size in I/R group (41.32±2.08)% was significantly increased (p<0.05). Compared with the I/R group, adenosine group (23.59±2.15)% and adenosine with lidocaine group (20.81±3.02)% significantly reduced the infarct size (p<0.05), and adenosine with lidocaine was better than adenosine (p<0.05).2. The detection of SOD activity:Compared with the sham-operation group (98.93±13.70) U/ml, the I/R group (50.61±12.58) U/ml was significantly reduced (p<0.05); Compared with the I/R group, adenosine group (68.05±10.53) U/ml and adenosine with lidocaine group (75.82±11.41) U/ml was significantly increased (p<0.05), and adenosine with lidocaine group was higher than adenosine group (p<0.05).3. The detection of MDA concentration:Compared with the sham-operation group (1.94±0.82) mmol/ml, the I/R group (8.63±1.97) mmol/ml was significantly increased (p<0.05); Compared with the I/R group, adenosine group (6.30±2.03) mmol/ml and adenosine with lidocaine group (4.05±1.89) mmol/ml was significantly reduced (p<0.05), and adenosine with lidocaine group was lower than adenosine group (p<0.05).Conclusion:Adenosine pretreatment reduces myocardial infarct size, increase serum SOD, decrease MDA content, effectively prevent MIRI, the effect of adenosine with lidocaine more effective.
