Study On Constituent From Polygala Chinensis L. And Optimum Preparation Process Of Polygalitol

Application of Chinese herbal medicine treatment has a long history in China, the current terms of the overall effect of Chinese herbal medicine treatment has increasingly presented a unique advantage. Now, China has twenty kinds of Chinese herbal medicine preparation, which included a pure Chinese herbal medicine and it is also added other constituents of Chinese herbal medicine-based formulations. Formulation have granules, tablets, oral liquid and so on.Polygala telephioides WILLD. have been show more precise treatment effect. The relational new drugs are being approvalled (or have been approwalled). The clinical and long-standing drug habit on traditional Chinese medicine, Polygala chinensis L. and Polygala telephioides WILLLD. are used as traditional Chinese medicine Polygala glomerata Lour. together. Today, we have few studies about Polygala chinensis L..We do not have find the report about constituents and active investigation. We have first studied the constituents of Polygala chinensis L. in order to expand the medicine resources.In this experiment, we have done the ethanol extraction of Polygala chinensis L. Throught macroporous resin, silica gel, reversed phase silica gel, Sephadex LH-20 we have isolated four compounds PC-1, PC-2, PC-3, PC-4. We identified these compounds thronght 1H-NMR,13C-NMR,HMQC,HMBC,MS spectrum and so on. These compounds was isolated from Polygala chinensis L. for the first time.Recently some research trends show that the contents of polygalitol in Polygala chinensis L. associated with the effect of drug dependence. So polygalitol is considered the effective constituents of Polygala chinensis L. The relational pharmacodynamic experiment is proceeding. So we have selected the extraction and purification process of polygalitol. At the same time, we have done optimized for the key steps.In the process screening experiments of polygalitol, we have tried some methods which included extraction, alcohol precipitation, column chromatography and so on. In the end, we determined the extraction process of Polygala chinensis L. through macroporous resin, silica gel column chromatography. The content of polygalitol that we obtained was 99.8%,melting point was 143-144℃,specific rotation was +41.58°,yield was 1.73%. This process is simple and the materials are easy to get,so it is can be used for mass production.
