| Objective: To study unsaturated fatty acids, hydrolytic amino acids of protein, unknown chemical constituents in the seed of stizolobium cochinchinensis.Methods: The unsaturated fatty acids were extracted by ligarine and analysised by gas chromatogram; sodium hydroxide solution and hydrochloric acid were used to extract and precipitate the protein, amino acids were analysised by amino acid analyzer; Ion exchange chromatography and silica gel column chromatography were applied to separate the unknown chemical constituents in the seed of stizolobium cochinchinensis, the structures were confirmed by Ultraviolet Spectrum, Infrared Spectrum, 1H-and13C-Nuclear Magnetic Resonance, Mass Spectrum.Results: Analytic results of the unsaturated fatty acids under area normalization method are: oleic acid 18.3%, linoleic acid 48.0%, linolenic acid 7.7%, these three unsaturated fatty acids account for 74.0%; Analytic result of amino acids hydrolizated from protein in Mucuna Cochinchinensis amounts to 25.8%, analytic result of amino acids hydrolizated from protein in residues of Mucuna Cochinchinensis is 21.2% in total; Two compounds are isolated, one is 5-((hydroxyl amino) methyl)-4-methyl-3-oxo-3,4-dihydropyrazine-2-carbimidic acid, the other is 2-(3,4-dihydroxy-2,5-bis(hydroxyl methyl) tetrahydrofuran -2-yloxy)-6-(hydroxyl methyl) tetrahydro-2H-pyran -3,4,5-triol(sucrose) based on the analysis of UV, IR, NMR ,MS spectrums.Conclusion: A certain amount of unsaturated fatty acids are detected in Mucuna Cochinchinensis, including oleic acid, linoleic acid and linolenic acid; the proportions of common amino acids hydrolizated from Mucuna Cochinchinensis and residues of Mucuna Cochinchinensis are 25.8% and 21.2%; A new compound is found named 5-((hydroxyl amino) methyl) -4-methyl -3-oxo-3, 4-dihydropyrazine-2-carbimidic acid, sucrose is isolated from Mucuna Cochinchinensis for the first time. |
