| Subject:By studying the technology of purificating total alkaloids in Ramulus Uncariae cun Unci by acidic exchange ion resin,select the optimum extraction process of the two kinds of alkaloids,and study the industrial preparation process of total alkaloids of TCM.Methods:Selecting Ramulus Uncariae cun Unci which mainly has low-polarity alkaloids as research objects,regarding rhynchophyline and total alkaloids in Ramulus Uncariae cun Unci as index, the study researched the optimum extracting process by orthogonal design,and selected the optional factors of absorption such as Resin models, Resin crosslinking, Resin column height and column diameter ratio, The column method, The column flow rate by single factor exploration, especially compared the eluting effect of different eluting methods with different eluting agent such as ammonia water(3-15%),10%ammonia water(contain 25-75% alcohol),hydrochloride solution(1 15%),5%hydrochloride solution(contain 25-75% alcohol), salt water(5-15%),salt water(contain 30-70% alcohol). and in the end selected the optional process.Result:leonuri was extracted with acid water,and then passed through the ion resins column(001×7,r:h=1:8)at the rate of 4BV/h in reverse direction,then the resin column was flushed with water at the rate of 6BV/h to pH6-7 in forward direction.At last, the column was eluted at the rate of 6BV/h to obtain alkaloids respectively with 5% salt water(contain 50% alcohol), the purification ratio of rhynchophyline and Isorhynchophylline is 92.48%, total alkaloid 92.77%.Conclusion:The technology of salt water eluting alkaloids in Ramulus Uncariae cun Unci by strong acidic exchange resins can achieve satisfactory results,and show the technology could overcome the defects such as low elution ratio, poisonous organic solvent and impurity, unsuitable for industrial production. The technology expands the scope of elluting agent,and has wide application prospects. |
