| Objective:By researching the effect of Di Huang Yin Zi(DHYZ) on the learning and memory ability in the place navigation test,neuropathology, oxidative stress of senescence accelerated mouse prone 8(SAMP8),investigate the mechanism and action of Di Huang Yin Zi on treating Alzheimer's disease.Methods:All the mice were male and 7 months'old.They have the same strain.SAMP8 were randomly divided into model group,DHYZ group,and Donepezil group.SAMP8'control group was senescence accelerated mouse resistance 1(SAMR1),the nomal group.The Morris water maze was applied to check these mice's learning and memory ability in the place navigation test. Enzyme activity of SOD,GSH-Px and content of MDA were measured by the method of biochemistry.HE staining was used to observe hippocampus's morphology and structure in the area of CA1.Results:1.SAMP8 showed the lower learning and memory ability in the place navigation test.In its liver, the enzyme activity of SOD and GSH-Px decreased, and the content of MDA increased.In its brain, the amount of pyramidal cells in hippocampus'area of CA1 decreased remarkably, the morphology and structure of cells were changed.2.DHYZ could shorten the escape latencies of SAMP8 remarkably in the place navigation test,and improve the learning and memory disorder of SAMP8.3.In the livers of SAMP8,DHYZ could increase the enzyme activity of SOD and GSH-Px,decrease the content of MDA,enhance SAMP8's anti-oxidative ability,and reduce the amount of lipid peroxides.DHYZ could improve the SAMP8's learning-memory ability.4.DHYZ could improve the cognitive ability of SAMP8 on changing the morphology and structure in the area of CA1.Conclusions: 1.SAMP8 is an ideal animal model of AD.2.It has a considerable improvement on the learning-memory ability in the place navigation test of SAMP8 with DHYZ.3.DHYZ could enhance the anti- oxidative ability of SAMP8.4.DHYZ could improve the degenerative process of hippocampus's morphology and structure in the area of CA1. |
