The Influence And Significance Of Sini Decoction' Pre-intervention On Experimental Hyperlipidemia Combined With Atherosclerosis In Rabbit Model

Objective:To observe the effect on TC,TG,HDL,LDL,Apo-A,Apo-B,Endothelin,Carbon monoxide and the changes in aortic endothelial plaque structures of HLP combined AS rabbits treated by Sini Decoction and to analyze the anti-hyperlipemia with atherosclerosis rationale of the therapy.Method:While fed with high fat food everyday and injected with BSA, egg-white protein, HLP combined AS rabbits were treated with different way of Atorvastatin and three doses of Sini Decoction(High, middle and low). To observe the changes after the intervention of the use of enzymatic determination of serum TC, TG, NO level; colorimetric assay after the intervention of blood LDL-CH, HDL-CH levels; electrophoresis after the intervention measured blood levels of apolipoprotein A and B changes; measured by radioimmunoassay in blood ET levels change; in situ hybridization measurement of MMP-2,9 expression; digital microscopic observation of rabbit aortic endothelial microstructure changes.Results:(1)On the area of increased NO levels in the blood, reduced the ET/NO ratio, aortic atherosclerosis MMP-9 expression and aortic atherosclerotic plaque thickness, while each SNT dose group compared with the model group, the value of NO is higher, but the value of ET/NO ratio, the expression of MMP-9 and the thickness of atherosclerotic plaques are lower. The difference between model group and each SNT doses has statistically significant (P<0.05 or P<0.01).(2)In reducing the levels of blood TG and atherosclerotic plaque area, the SNT low-dose group's value were less than model group, but the difference has not statistically significant (P>0.05); in the meanwhile the values of SNT middle and high doses group were less than the model group, the difference has significant (P<0.05 or P<0.01).(3)In reducing blood TC, LDL, Apo-B, ET levels, ET/NO ratio, the expression of MMP-2 in aortic atherosclerotic plaques, SNT low and medium dose group's levels were less than the model group's, but the difference was not significant (P>0.05). The levels of SNT high-dose group were less than too, the differences have obviously significations (P<0.01). In elevating levels of blood's HDL and Apo-A contain, HDL/LDL and Apo-A/Apo-B ratio, The values of each SNT dose group were higher than the model group, while the differences of model group and SNT low-dose group or SNT medium dose group, have no significant difference (P>0.05), the difference between SNT high dose group and model group was statistically significant (P<0.01).Conclusion:(1) SNT can adjust the lipid metabolism of experimental hyperlipidemia combined with atherosclerosis in rabbit model, lower cholesterol and triglyceride levels and decreased serum low density lipoprotein and apolipoprotein B levels, so that high-density lipoprotein, apolipoprotein A increased.(2)SNT has increased NO content in blood, reduce content of plasma ET, and inhibited the expression of matrix metalloproteinase 2,9 in rabbit aortic atherosclerotic plaques, played a role in reducing the formation of AS plaque.(3)SNT to prevent the formation mechanism of AS may be related to blood lipid metabolism, aortic endothelial damage, slow down the lipid granules. SNT may inhibit gene expression, enhance antioxidant enzyme activity and stability of atherosclerotic plaque, thereby enhancing the aortic wall antioxidant, anti-damage and reduce lipid particles in atherosclerotic plaque deposition and off, thus slowing the further development of AS, to prevent serious complications.
