Effects Of Yanghe Decoction Containing Serum On Osteoblast And Osteocblast

Objective To investigate the effect of Yanghe decoction On anti-tumor activity and osteoblas,osteoclast by serologic pharmacological method and provide theory basis for Yanghe decoction to popularize and apply in clinic practice.It also provides the theory basis for warm-method treating tumor of traditional Chinese medicine.METHODS 1. Randomly grouped 20 SD rats, intragastric administration, obtained low dose serum of the Yanghe decoction,middle'dose serum of the Yanghe decoction high dose serum of the Yanghe decoction,and free medicine.2. dissociation the OB of rats.cultivate the cell of UMR106 OB,RAW264.7 and Inducible factor M-CSF and RANK-L induced osteoclast formation from RAW264.7 cells.3.MTT assay is used to calculate the inhibitory rate of UMR106,OB,RAW264.7 and osteoclasts (TRAP) enzyme activity. Results 1. Drug-containing serum was successfully gained, and was administrated on the OB,OC,cells line 106 and RAW264.72. OB, cells line UMR106, RAW264.7 were successfully cultivated. OC was also differentiation from cells line RAW264.73. Via method of MTT, value of OB was related to the concentration of drug-containing serum and treating time. Serum containing high dose and medium dose of Yanghe decoction can remarkablly improve OB value in 24h,48h, as well as 72h. While low dose of it can slightly promote the OB value only on 48h and 72h, but the data was out of significance. Same as it is, all the concentration of Yanghe decoction serum can improve the OB value when they were administrated in cells line UMR106, the significance were remarkable on 24h,48h, and 72h. On the other hand, all the concentration of Yanghe decoction serum drop the OB value of cells line RAW264.7, but was out of significance compared to control serum. Along with time, OB value was normally rise, which indicated that the cells were soundly alive. Serum containg Yanghe decoction can not prohibit the differentiation of OC from RAW264.7, which was a result from TRAP test.Conclusion Results from differentiation of OB, cells line UMR106, RAW264.7 and differentiation of OC have demostrated that, the Yanghe decoction containing serum could significantly improve the proliferation of OB, while the prohibiting effect on OC was out of statistics significance. In addition, the prohibiting effecton differentiation of OC was neither remarkable.
