| Malignant tumor is a cell active proliferating abnormality disease,has been seriously threatening human being's life and health, it inevitably to lead to death without control.Now,the treatment to it is the synthetic application of surgery, chemotherapy,radiotherapy and biotherapy. although some patients on early stages have a chance to cure,but because of many kinds of reasons,many patients have been belonged to advanced stage when discover,miss to operate. The chemotherapy has an influential position in the treatment of the patients who are in the advanced stage and have definite efficacy, but they also have some problems for their severe side effects, high price and multidrug resistance, these can lead to the aggravation of patients' conditions and the degression of quality of life, even lead the patients to give up. As is know to all tumor is a anvanced, very long process,obtain the opportunity of thorough radical is rarely. But we hope can enhance the immune function of organism maximatily, control and stable tumor,diminished harm of tumor, make the patients keep a certain work capability and a better quality of life long-term, the existence of band-tumor for long time has been our reality strive target. So, we need research one convenient feasible medicines or methods, which not only to prevent or slow down the growth of tumor, and also safely, can long-term even follow lifetime. Traditional Chinese medicines treat tumors in a systemic way,have their merits such as widespread medicinal herbs resource,low price and little side effects.In recent years,traditional Chinese medicine, which is effective in the combined therapy of malignant tumor,has been accepted by more and more experts and patients.Yiliuning is a clinical experienced prescripsion of our technical office,is a traditional Chinese compound prescription composed of Rab,Aca, Duc and Sol.It has definite curative effect on the treatment of tumours of digestive system,and has little side effects.According to our experimental result of its four compositions' extracts: Rab compound with Aca, Duc and Sol can arrest the Eca-109 cells during S-phase and induce cell apoptosis; the mouse's acute toxicity experiments show its maximatily dosage is 126 times than clinical;and the pharmacodynamics experiment has confirmed that the combined use of four drug's tumor inhibition rate greater than 30% to H-22 and S180, and can extend survival days of leukemia L1210 tumor-bearing mice.The objectives of this experiment are to observe the effect of Yiliuning's anti-tumor and potential mechanism of action on Ec-9706 bearing nude mouse, provide experiment basis for the use of clinic.Methods:The experiment of anti-tumor on Ec-9706 bearing nude mouse and immunityThe model of nude mice bearing tumor were established by inoculating Ec-9706 cells.tumor bearing nude mice were randomly divided into five groups and each group consisted of seven nude mice:Negative control group(the same volume of physiological saline was affused by mouth);Positive control group (through peritoneal injection PYM 4mg/kg.d, three times/week);high, moderate, low three dosage group of Yiliuning(6.64g/kg.d,13.28g/kg.d,19.92g/kg.d of Yiliuning'extracts were respectively affused by mouth);After giving drugs for four weeks continuously,from 7d observe the change of tumor's weight and vital sign of nude mouse.29d execute moses, we took the neoplasm to weigh,computed the rate of inhibiting tumor and at the same time make paraffin blockpathological section, use the immunity to detect every group's cox-2 and ki-67's expression. Took liver,pancreas,spleen,kidney through HE dye observe organ's cellpathology under the optical microscope, understand the toxic effect of drugs to organs.Results:The tumor inhibition ratios of groups of Yiliuning with small,moderate,high dosage are 35.13%,35.09%,35.59%respectivly, while that of positive control group is 43.40%,Compared with negative control group,IR difference of all groups are statistically significant:the groups of positive control group and Yiliuning with small,moderate, high dosage and (P<0.01),The Yiliuning groups have no apparently dose-effect relationship.when the experiment is over,the positive group and yiliuning group's weight all higher than the negative control group.The results of pathomorphology indicate that there are Yiliuning have no obviously side effect to organs of mouse.The expression frequency of cox-2 and ki-67 of Yiliuning groups and positive control groups are lower than negative control group.Compared with the negative control group, the difference of the Yiliuning groups and positive group are statistically significant:the small dosage (P<0.05),the moderate and high dosage (P <0.01).Conclusion:1.Yiliuning can improve obviously body weight and general state of health of the nude mouse bearing Ec-9706.2.Yiliuning can resist the growth of tumor of the nude mouse bearing Ec-9706, and can promote neorobiosis and apoptosis of tumour cell.3.The mechnism may change the expression of cox-2 and ki-67 to prevent the cell splitting, so as to inhibit the growth of tumor cells. |
