Objective To investigate the effects of simvastatin and losartan on the expression of Toll-like receptor 4 and NF-κB in the arteries of diabetic rats.Method 54 SD rats were randomly divided into control group (A group, n=6), diabetic group (B group, n=12), simvastatin group (C group, n=12), losartan group (D group, n=12) and the combined treatment group (E group, n=12). Respectively,8 weeks,12 weeks to take half of each group were anesthetized rats were sacrificed, weighed, cut open the chest, heart puncture blood, blood glucose, insulin and lipids (including TC, TG, LDL-C, HDL-C.) Light carotid artery morphological changes were observed, the use of BI-2000 image analysis system for HE staining and immunohistochemical image analysis slices carotid artery intima thickness and optical density analysis of the arterial tissue of rats TLR4 and NF-κB expression, reverse transcription polymerase chain reaction (RT-PCR) for quantitative analysis.Results(1)Diabetic rats had significantly "a little" symptoms and significantly increased blood glucose, serum insulin levels were significantly lower, no significant effect of drug intervention;(2)Markedly abnormal blood lipids in diabetic rats, simvastatin group and the combined treatment group to reduce TC, TG, LDL-C expression, little effect on losartan potassium;(3)Light microscope with the time arrangement of vascular endothelial cells in diabetic rats gradually disorder, obvious proliferation of medial smooth muscle cells, or even foam cells, resulting in carotid artery intima thickening, stenosis, simvastatin intervention group and significantly improve the effect of the combination group, losartan intervention group improved the general effect;(4)Immunohistochemistry results showed that:compared with the normal control group, diabetes group, TLR4 and NF-κB expression was significantly increased expression decreased after drug intervention, the most obvious combination group, followed by simvastatin group, losartan the worst of potassium intervention group;(5) Quantitative PCR analysis further corroborated the results of immunohistochemistry, and the difference is more obvious;(6) In the diabetes group, TLR4 and NF-κB expression with intima media thickness was positively correlated.Conclution This study shows that diabetic rat carotid artery tissues TLR4/NF-κB expression and diabetic rat carotid artery atherosclerosis in the genesis and development are closely related. Can be TLR4/NF-κB macrovascular disease as early detection indicators. Simvastatin and losartan potassium of diabetic rats, respectively, through the carotid artery tissue TLR4/NF-κB expression, inhibition of vascular endothelial inflammatory response, vascular smooth muscle cell migration, proliferation and foam cell formation, reduce the pathological carotid artery changes, delayed carotid atherosclerosis process, which play a protective role in artery, and the former is stronger, both more obvious when combined.
