This paper mainly discussed the cancer prevention effect and antitumor effect of isothiocyanates(ITCs); The experiment firstly optimized the extraction process of the volatile oil from Semen Sinapis Albae and analyzed the chemical components of the volatile oil, then elaborated the antitumor effect of volatile oil from Semen Sinapis Albae on H22 cells and H22-bearing mice. And the mechanism of anti-tumor effect was investigated, which might provide better thoughts of using the volatile oil from Semen Sinapis Albae.The optimum extraction condition was investigated by the orthogonal design L9)(34), and the chemical components of the volatile oil from Semen Sinapis Albae were analyzed by GC-MS;the inbibitional effect of the volatile oil from Semen Sinapis Albae to H22 cells was detected by Trypan blue staining and MTT assay in vitro;the influence of H22 cells'apoptosis and cell cycle by the volatile oil was detected by DNA agarose gel electrophoresis and Propidium iodide(PI) staining; Ascites tumor model and solid tumor model was used to detect the inbibitional effect of the volatile oil from Semen Sinapis Albae to H22-bearing mice in vivo, the morphologic changes of the tumor cells was observed by Hematoxylin-eosin(HE) staining, spleen index and thymus index was detected to investigate the toxicity and side effects of the volatile oil from Semen Sinapis Albae, and the acridine orange(AO) staining was used to observe the morphologic changes induced by apoptosis, the expressions of the gene Bax and Bcl-2 in the tumor tissue was determined using immunohistochemistry.The results show that the optimum extraction conditions of the volatile oil from Semen Sinapis Albae is:Hydrolysis temperature 50℃, pH value of buffer solution 5.0, concentration of ascorbic acid 0.5mmol/L,the yield rate of the volatile oil of this process can reach 0.164%, the main component of the volatile oil is allyl isothiocyannte(89.411%); the volatile oil from Semen Sinapis Albae can inhibit the growth of H22 tumor cells significantly in vitro and it may work through apoptosis; the volatile oil from Semen Sinapis Albae also can inhibit tumor growth in vivo and extend the life span of H22-bearing mice significantly,and the results demonstrated a good dose-effect relationship, but the high-dosage group of the volatile oil has obvious toxicity and side effects on the mice; the volatile oil from Semen Sinapis Albae may raise the expression of Bax and suppress the expression of Bcl-2,as a result,raise the Bax/Bcl-2 rate to promote apoptosis.
