Objetive: To observe different blood serum content of radix astragali in Spiritual cultivates Decoction in impacting cell proliferation of H2O2 inducing Human umbilical vein endothelial cell(HUVEC) oxidative damage model and protecting HUVEC damage.And to discuss accurate mechanism of Spiritual cultivates Decoction in treatment of unstable angina pectoris.To offer reference for further research and development.Method:The thirty vistar rats were classified into 2 groups at random, radix astragali group and blank group. Make 1.03g/ml drug components of radix astragali(doses= Commonly used quantity×animal equivalent area coefficient×5), blank group rats were given isodose normal saline to lavage, 2 times a day, morning and evening, for one week, to sample heart boold at last lavage after 1 hours and centrifugalize for prepare. Freezing HUVEC was rivived , then cultivated and passaged. To cultivate HUVEC and different blood serum content of radix astragali(2%,4%,6%).Experiment was divided into 5 group:Blank blood serum group: To cultivate for 26 hours in blank blood serum nutrient solution.model group: To cultivate for 24 hours in basal nutrient solution, adding 100μmol·L- 1 H2O2 for two hors.2%,4%,6%blood serum content of radix astragali group: To cultivate for 24 hours in basal nutrient solution of 2%,4%,6% blood serum content of radix astragali , then adding H2O2 for two hors.Different blood serum content of radix astragali could influence the change of HUVEC growth activeness through through the MTT and morphology method observation. Rusult:The statistics show that cell OD in 2%,4%,6% blood serum content of radix astragali group were higher than H2O2 model group. There had statistical significance between three groups and H2O2 model group(p<0.01). 4% blood serum content of radix astragali group was obvious.But 6% group was most significant. The difference had statistical significance between three groups. Morphology observe:2% blood serum content of radix astragali group:cell volume becomed smaller,gap larges obviously , relates each other less,cell drops obviously,cell deformation and shrinking are improved.4% blood serum content of radix astragali group:cell volume becomed smaller,gap become larger, relates each other, part of cell drops, cell deformation and shrinking are improved the most obviously.6% blood serum content of radix astragali group:cell volume becomed smaller,gap become larger,relates each other,samll part of cell drops , cell deformation and shrinking are improved obviously.Conclusion: It was found that blood serum of radix astragali could promote cell proliferation of HUVEC at normal cultured state, multiplication effect strengthen with the increase of blood serum concentration of radix astragali.Compared to control group, It could increase vessel endothelial cell activation and promote cell metabolism proliferation.Infer:through promoting cell multiplication, radix astragali could increase vessel endothelial cell activation, adjust constriction of blood vessels,had the anti-thrombosis effect.And it could prevent or delay initiation of atherosclerosis, prevent and treat atherosclerosis caused by the injury of vessel endothelial cell.It might be one of function mechanisms prevtenting unstable angina pectoris.
