| Objective: To investigate the influence of prepared radix rehmanniae polysaccharides(RPS) on mice chromosomal mutagenic induced by CTX, mechanism of antitumor effect of RPS. To provide a scientific basis for the chinese medicine development, utilization and clinical safe application.Methods: To gain the RPS by water extraction and alcohol precipitation. To evaluate the antimutagenic effects of RPS by mouse bone marrow PCE micronucleus test, CA test and SCE test. Determination of in vivo experiments using Rehmannia glutinosa polyses on the S180,H22 tumor-bearing mice in-hibited tumor; To detect the RPS on tumor tissue and serum protein expression patterns of S180 tumor-bearing mice by SDS-PAGE.Results: RPS has a good inhibitory effect on CTX-induced mouse bone marrow PCE micronucleus, CA and SCE. RPS medium dose group has the best inhibitory effect, the inhibition rates were: 27.96%, 32.04%, 25.61%. The re-suct indicateel thet RPS has the antimutagenic function. RPS on the S180, H22 tumor-bearing mice inhibited tumor in vivo results show that RPS can inhibite the S180, H22 tumor. The medium dose group has best inhibitory effet, the in-hibition rates were 36.36%, 42.76%. RPS can extend S180, H22 tumor-bearing nice survival time. RPS on S180 tumor-bearing mices tumor tissue protein and serum protein expression showed. RPS can improve the expression of tumor protein, make its expression was similar to the normal control group, After administration, compared with other group the RPS group expressed another protein component which molecular weight is 16KD. Make the mice protein serum on the molecular weight of 40KD, 14.4 KD reducted, molecular weight of 26 KD increaseel, There is no significant difference compared with the normal group.Conclusion: RPS on CTX-induced micronuclei in mice bone marrow PCE significantly inhibited; on CTX-induced bone marrow cells in mice sig-nificantly inhibited the rate of CA; On CTX-induced SCE rate in mouse bone marrow cells inhibited; RPS can inhibit S180, H22 tumor-bearing mice tumor growth; RPS to extend S180, H22 survival time of mice; RPS to S180 tu-mor-bearing mice on tumor tissue and serum of a protein with the impact of some. |
