| Ganoderma lucidum is a famous medicinal fungi, its main chemical components of Ganoderma lucidum polysaccharides, Ganoderma lucidum triterpenes. Therefore, The study has important theoretical and practical value. It includes reseraching the variation regulation of polysaccharides and triterpenoids in the liquid fermentation phase and the growth phase, and the extraction methods of fruiting body polysaccharides,and the conditions of artificial cultivation and liquid fermentation.The method is finalized that microwave is the most suitable method of Ganoderma lucidum polysaccharides, through comparative studies of different extraction methods of polysaccharides, the optimum process parameters are: soak time 1.5 h, microwave treatment time 15 min, solid-liquid ratio 1:110, pH 8.0.Under these conditions, Ganoderma lucidum polysaccharides reach 32.001 mg / g.The changes of polysaccharide and triterpenoids in growth process of Ganoderma lucidum , the culture substrate and form are studied. The optimal cultivation pattern of Ganoderma lucidum is to cultivate with sawdust in shed .Polysaccharide content is higher in budding time , slight decline in beginning period , rapid increasing in parachute period , reaching the highest in maturity phase and slightly lower after spore ejection. The content of triterpenoids is in the stable, only has a slight decrease after spore ejection.By experiment on single factor, the optimal condition of Ganoderma lucidum fermentation is as follow: glucose 2 %, peptone0.5 %, VB110mg/L, KH2PO4 0.1 %, MgSO4·7H2O 0.05 %, pH 5.5, temperature 28℃, culture time 120 h. Under these condition, biomass reaches 44.548 g/100 mL. The highest content of polysaccharides appears at 144 h reaching 2.452 g/100 mL and the highest content of triterpenoids appears at 168 h reaching 0.274 g/100 mL. |
