Lysimachia clethroides Duby is a perennial herb indigenous to many parts of the tropics and sub-tropics, including east and south China regions. The herbal medicine, Lysimachia clethroides Duby, has long held a place in natural medicine in the sub-tropical regions where it grows. People in east and south China regions use Lysimachia clethroides Duby for edema, pyretic stranguria, jaundice, furuncle and leukemia in folklore. Previous studies on constituents from the Lysimachia genus led to the isolation of different compounds such as flavones, triterpenoids and benzoquinones.In the course of our investigation for bioactive agents from the antitumor active fraction of Lysimachia clethroides Duby, 14 compounds were obtained by using silica gel column chromatography, Sephadex LH-20 chromatography, MPLC, and Semi-HPLC, and structures of 14 compounds were elucidated through spectroscopic analysis (NMR, MS and IR ). Their structures were identified to be dihydrokaempferol (ZW-1), naringenin (ZW-2), eriodictyol (ZW-3), kaempferol (ZW-4), quercetin (ZW-5), (-)-epicatechin (ZW-6), prunin (ZW-7), isoquercitrin (ZW-8), kaempferol-7-O-β-D-glucoside (ZW-9), kaempferol-3-O-β-D-rutinoside (ZW-10), isorhamnetin-3-O-β-D-rutinoside (ZW-11), rutin (ZW-12), kaempferol-3-O-(2, 6-dirhamnopyranosyl)glupyranside (ZW-13), quercetin-3-O-(2, 6-dirhamnopyran osyl)glupyranside (ZW-14). Among them, compounds ZW-1, ZW-3 and ZW-14 were obtained from this genus for the first time.Based on the studies of chemical constituents, five flavonoids (kaempferol, quercetin, prunin, isorhamnetin-3-O-β-D-rutinoside, rutin) were selected as marker component, and their contents in the antitumor active fraction of Lysimachia clethroides Duby were determined by using HPLC with dual wavelength detection. The validated method is simple, efficient, and able to provide the basis for the quality control of flavonols in active fraction of Lysimachia clethroides Duby.
