| 0bjective To explore the expression of livin gene in acute lymphocytic leukemia (ALL ) cells and its clinical significance. Observes the Livin gene the expression or not and the expression relative quantity height to the acute adult ALL morbidity,the recrudescence, whether there is the prognosis relevance. And analyze the relationship of between the clinical characteristics and the gene expression of Livin .Materials and Methods (1) Object of study is adult acute lymphoblastic leukemia patients in 52 cases that were untreated or relapsed from August 2008 to June 2009,the First Affiliated Hospital of Anhui Medical University. normal control group was 12 normal bone marrow smears in patients with non-malignant hematological disease.(2) The mRNA expression of livin gene in 52 ALL adult patients(including denovo acute lymphocytic leukemia patients,relapsed patients and complete remission patients) ,were measured by using reverse transcription polymerase chain reaction (RT-PCR) . Other 12 healthy adults we reselected as normal controls (NC ) .Take looks after the house the geneβ-actin to illuminate as the materials for internal reference.(3) SPSS13.0 statistical software was used for statistical analysis.Results The results showed that the mRNA rate of livin gene in ALL patients was significantly higher than that in NC (P < 0. 01), while it decreased in patients with complete remission (CR) (P < 0. 05); In relapsed patients, the expression of livin mRNA increased again. High-risk group Livin gene mRNA expression level is higher than standard risk group ,differences between two groups was statistically significant (p <0.01). In the revisit observation initially governs with recrudescence adult ALL patient, the CR rate of patients with livin positive was lower than that of patients with livin negative (P < 0. 05) . Livin mRNA expression masculine patient the position life may short in the negative expression patient. There is a positive correlation between Livin mRNA expression levels in adult ALL patients with peripheral blood white blood cell count (r = 0.815, P = 0.006).Conclusion It is concluded that expression of livin gene may play a synergic role in the pathogenesis of ALL and associates with CR rate in ALL. It seems that expression of livin gene may be used as a marker of poor prognosis in adult acute lymphocytic leukemia. Expression of livin gene have associations with risk group and response to chemotherapy, while have not associations with patient sex and immunophenotype. |
