Purpose1. To study the effects of Leonurine (Leo), Stachydrine (Sta) Leo+Sta on acute myocardial ischemic in mice.2. To evaluate the effects of Leo, Sta, Leo+Sta on isolated rat spontaneous and spasm uterus.3. To study the mechanism of Leo, Sta, Leo+Sta on isolated rat spasm uterus.4. To study the effects of Leo, Sta, Leo+Sta on membrane potentials in mice smooth muscle uterin cell.Methods1. The acute myocardial ischemic model was made by using subcutaneous injection of isoproterenol ( Iso) in mice. T wave of electrocardiogram ( ECG) was observed, the activitise of lactate dehydrogenase (LDH) and the contents of malonaldehyde (MDA) in serum were measured, Jin′s formula was applied to analyze the combination of Leo and Sta, The histopathological changes of myocardium were observed by using HE staining.2. Measurement of isolated uterine contractions. To observe the changes of the frequency, amplitude, uteri activity in spontaneous contractions and spasm contractions induced by oxytocin. 3. After preperfusion of L-NAME or Indo, the effects of Leo, Sta, Leo+Sta was observed on frequency in spasm isolated rat uterus.4. Measurement of membrane potentials in mice smooth muscle uterin cell.Results1. Both the combination Leo 2.5 mg/kgwith Sta 5 mg/kg and Leo 5 mg/kgwith Sta 10 mg/kg obviously inhibited the changes of T wave induced by Iso, besides the effects of the inhibition continued to 90 min. Using Jin′s formula to calculate the value of q between 0.85 and 2.0, effects of these combination on the changes of T wave were simple combined or enhanced. They obviously reduced the contents of MDA and the activities of LDH in serum, in comparison with Leo 5 mg/kgor Sta 10 mg/kg alone, the combination of Leo 5 mg/kg and Sta 10 mg/kg was remarkably less in the contents of MDA and the activities of LDH. compared with Leo and Sta, they significantly ameliorated myocardial ischemic injury, only few constriction bands of heart muscle fiber were seen.2. Leo (10-5M) could increase the frequency and decreased the amplitude of spontaneous contractions in the rat uterus, Sta could reduced the frequency and uteri activity, Leo+Sta (10-6, 10-4M) could reduced the frequency.3. Leo could reduce the frequency and amplitude of spasm contractions in the rat uterus, Sta could reduced the frequency, amplitude and uteri activity, Leo+Sta could reduced the frequency and amplitude.4. After preperfusion of L-NAME, the reduction of frequency induced by Leo, Sta, Leo+Sta was inhibited in spasm uterus contractions.5. After preperfusion of Indo, the reduction was not obviously effected in spasm uterus contractions.6. Leo, Sta, Leo+Sta could lead cells produce hyperpolarzation in mice smooth muscle uterus, while hyperpolarzation produced by Leo related with concentration. ConclusionsIn summary, Leo and Sta combined had more resistant effects than Leo or Sta alone on acute myocardial ischemic, Leo had bilateral effects on spontaneous contractions. Sta, Leo+Sta could relax spasm contractions, NO might be involved in the mechanism of the relaxation activity of Leo, Sta, Leo+Sta on spasm contractions, while PGs might not be involved. Our study may provide a certain reference value for clinical application of Leo, Sta and Leo+Sta in the treatment of dysmenorrhoea and myocardial ischemia.
