Objective:To investigate the effects of adiponectin on proliferation and the expression of VEGF and reveal the probable protection mechanism for HCM of adiponectin in D-glucose of high concentration by cultured with high concentration of D-glucose.Methods:â‘ The human cardiac myocyte (HCM) were divided into 3 groups randomly: normal glucose group (5.5 mmol/L glucose),high glucose group (30.0mmol/L glucose), and adiponectin group(30.0mmol/L glucose+2.5ug/ml adiponectin). After HCM was culturedwith upper three different kinds of environments for 24h,48h,72h.The proliferationactivity of human cardiac myocyte was observed by MTT.â‘¡The amount of VEGF mRNAwas observed by Real time PCR,after HCM was cultured with upper three different kindsof environments for 24h,48h,72h.Results:â‘ In the 24h high glucose group compared with normal HCM proliferationpromoted(P<0.05), while the high glucose group compared with adiponectin had no statisticaldifference (P>0.05).Compared with that in normal glucose group,HCM proliferation activitywas inhibited in high glucose group in 48h,72h(P<0.05).But the proliferation of HCM was increased after adiponectin being added(P<0.05),showing the characteristic of time dependence.â‘¡The expression of VEGF mRNA was significantly higher in high glucose group(P<0.05),but it was inhibited in adiponectin group compared with high glucose group. The differencesbetween the expression of VEGF at any time-point in high glucose group and in adiponectingroup were statistically significant(P<0.05),also showing the characteristic of timedependence.Conclusion:HCM stimulation can inhibit cell proliferation,induce apoptosis afterlong-term high glucose.The result indicate that adiponectin can promote the proliferation of the HCM and protect them in D-glucose of high concentration.
